تاثیر مدت زمان پیش تیمار آب مقطر بر شاخصهای جوانه زنی و بیوشیمیایی بذرهای زوال یافته کوشیا (Kochia scoparia L. Schard)

چکیده

زوال بذر پدیده­ای طبیعی است که بذرها قوه نامیه و کیفیت خود را حتی در شرایط مطلوب نگهداری از دست می­دهند. در این آزمایش تاثیر پیش­تیمار آب مقطر بذر در  بهبود خسارت ناشی از زوال بذر کوشیا بررسی شد. آزمایش در قالب طرح پایه کاملاً تصادفی در چهار تکرار انجام شد. بذرهای کوشیا به روش پیری تسریع شده به­مدت 48 ساعت در دمای 40 درجه سانتی­گراد زوال یافتند. سپس بذور زوال یافته در دمای 20 درجه سانتی­گراد به­مدت 5/1، 3 و 5/4 ساعت با آب مقطر پیش­تیمار شدند. نتایج نشان داد، پیش­تیمار آب مقطر در تمام زمان­ها به­طور معنی­داری موجب بهبود درصد جوانه­زنی، سرعت جوانه­زنی، طول گیاهچه، شاخص بنیه، فعالیت آنزیم­های آنتی اکسیدانی (کاتالاز و سوپراکسید دیسموتاز)، قندها و پروتئین­های محلول بذرهای زوال یافته گردید. متوسط زمان جوانه­زنی و محتوای مالون دی­آلدهید بذرهای پیش­تیمار شده در مقایسه با بذرهای  پیش­تیمار نشده کاهش یافت. پیش­تیمار آب مقطر به­مدت 5/1، 3 و 5/4 ساعت درصد جوانه­زنی  را 15، 23، 37، سرعت جوانه­زنی را 4/45، 7/72، 109 و فعالیت آنزیم سوپراکسید دیسموتاز را به ترتیب 6/8، 14، 27 درصد در مقایسه با بذرهای پیش­تیمار نشده افزایش داد. بنابراین، کاربرد آب مقطر به­مدت 5/4 ساعت به­عنوان زمان مناسب پیش­تیمار جهت بازیابی کیفیت از دست رفته بذرهای کوشیا و بهبود خصوصیات جوانه­زنی آن قابل توصیه می­باشد.

The deterioration of the seed is a natural phenomenon, and seeds tend to lose quality and viability even under optimal storage conditions. The ability of hydropriming to ameliorate seed deterioration damage was studied in kochia. The experiment was a completely randomized design with four replications. Kochia seeds were subjected to accelerated ageing for 48 hours at 40 °C and then hydroprimed at 20 °C for 1.5, 3 and 4.5 hours. Regardless of duration, hydropriming significantly improved final germination percentage, germination rate, seedling length, vigour index, antioxidant enzyme activities (catalase and superoxide dismutase), soluble sugars and proteins of aged seeds. Mean germination time and malondialdehyde content of primed seeds decreased compared to non-primed seeds. Hydropriming for 1.5, 3 and 4.5 hours increased final germination to 15, 23 and 37 %, germination rate by 45.4, 72.7 and 109 % and superoxide dismutase activityby 8.6, 14 and 27 % compared to non-primed seed, respectively. Thus, hydropriming for 4.5 hours is the most suitable priming period to recover loss of kochia seed quality and to improve germination characteristics of deteriorated seeds.
