بررسی تاثیر انبارداری و روش‌های مختلف پرایم نمودن بر صفات جوانه زنی نمونه‌های بذر گونهHalocnemum strobilaceum موجود در بانک ژن منابع طبیعی ایران

چکیده

سرعت زوال بذر و تاثیر روش‌های مختلف پرایم بر بهبود جوانه‌زنی بذور اکسشن‌های مختلف بذر گیاه Halocnemum strobilaceum  موجود در بانک ژن منابع طبیعی ایران در این آزمایش بررسی شد. برای بررسی سرعت زوال بذور، آزمون متداول جوانه‌‌زنی بر روی 42 اکسشن متفاوت از نظر محل و زمان جمع‌آوری، انجام گردید. تاثیر روش‌های مختلف پرایم شامل تیمارهای اسید جیبرلیک در دو سطح 250 و 500 میلی‌گرم در لیتر، پلی‌اتیلن‌گلایکول در دو سطح 6/0 و2/1 مگاپاسکال، نیترات‌پتاسیم با غلظت 78/19 میلی­مولار، سرمادهی مرطوب در دمای چهار درجه سلسیوس، به­مدت یک ماه، کلرید‌سدیم با غلظت 100 میلی­مولار بر روی سه اکسشن 1645، 33515 و 43882 (به­ترتیب 17، 9 و 5 سال در سردخانه نگهداری شده بودند) آزمایش شد. نتایج نشان داد میانگین سرعت زوال بذور 53/3 درصد به ازاء هر سال نگهداری در دمای چهار درجه سلسیوس می­باشد. همچنین واکنش بذور هر اکسشن به تیمارهای مختلف پرایم متفاوت است. به­طوری­که اکسشن 1645 بر اثر تیمار کلرید‌سدیم با غلظت 100 میلی­مولار و سرمادهی، اکسشن 33515 با تیمار اسید جیبرلیک با غلظت 500 میلی‌گرم در لیتر و اکسشن 43882 در شاهد، درصد جوانه‌زنی بیش­تری داشتند. در صفت طول ریشه­چه و ساقه­چه هیچ تفاوت معنی‌داری بین تیمارها مشاهده نشد. از نتایج این آزمایش می­توان استدلال نمود که تیمارهای پرایم فقط می­تواند توانایی ذاتی بذر را به مرحله ظهور برساند. لذا برای جمع‌آوری و ذخیره بذور برای مدت طولانی سلامت و کیفیت اولیه بذر باید مورد توجه قرار گیرد.

To evaluate the deterioration rate and effect of different priming methods on improving seed germination on different seed accessions of Halocnemum strobilaceum provided by Natural Resources Gene Bank of Iran, To evaluate the deterioration rate, a standard germination test was performed on 42 seed accessions with different origin and collection dates. To study effect of different priming methods, treatments included gibberellic acid (250 and 500 mg/l), polyethylene glycol (-0.6 and -1.2 MPa), potassium nitrate 19.78 mM, stratification at 4 °C for one month, NaCl 100 mM and Chilling were compared in 1645, 33515 and 43882 accessions That5, 9 and 17 years were stored in the refrigerator. The results showed that the average seed deterioration rate was 3.53 %/y of stored seeds at 4 °C.Each seed accession showed a different reaction to prime treatment. Results indicated germination percentage improved in the accession 1645 by treatment NaCl 100 mM, in the accession 33515 by treatment gibberellic acid 500 NaCl 100 mM, and in the accession 43882 by control treatment. However, no significant differences were observed between radicle and pedicle lengths among different treatments. From the results of this experiment, it can be concluded that priming treatments can only express the potential of the seed. Therefore, for collecting and storing seeds for a long time, the health and initial quality of seeds are very important.
