بررسی اثر پرایمینگ برجوانه‌زنی بذر و صفات مورفو-فیزیولوژیکی گیاه دارویی خرفه (Portulacaoleracea L.) تحت تنش شوری

چکیده

تحمل نمک در مرحله جوانه­زنی مهم می­باشد، زیرا افزایش شوری معمولاً در سطح خاک شدیدتر است. به­ویژه در مواردی که سطح آب زیرزمینی بالا باشد. به­منظور بررسی تأثیر پرایمینگ و تنش شوری بر صفات کمی و کیفی گیاه دارویی خرفه، این آزمایش در آزمایشگاه فناوری بذر دانشکده علوم کشاورزی دانشگاه شاهد در سال 1398 اجرا شد. عامل­های آزمایش شامل پیش تیمار (شاهد (آب مقطر)، هیدروپرایمینگ، نیترات پتاسیم و اسید سالیسیلیک) در واکنش به 6 سطح شوری عدم اعمال شوری به­عنوان شاهد و شوری30، 60، 90، 120 و 200 میلی­مولار با کلریدسدیم بودند. این آزمایش به­صورت فاکتوریل در قالب طرح پایه کاملاً تصادفی با سه تکرار مورد مطالعه قرار گرفت. صفات مورد اندازه­گیری شامل شاخص­های جوانه­زنی، رنگدانه­های فتوسنتزی، محتوای آنتوسیانین، پرولین و قندهای محلول بودند. نتایج نشان داد که بیش­ترین میزان مؤلفه­های جوانه­زنی همچون درصد جوانه­زنی (97درصد)، سرعت جوانه­زنی (16/2بذردرروز)، طول گیاهچه (69/6 سانتی­متر)، وزن تر گیاهچه (4گرم)، شاخص طولی بنیه گیاهچه (89/324) و محتوای نسبی آب (25/75 درصد) در عدم تنش شوری و کاربرد هیدروپرایمینگ به­دست آمد. در سطوح شوری 60 میلی­مولار، محتوای کلروفیل a، کلروفیل b و کلروفیل کل در تیمار هیدروپرایمینگ افزایش یافت و بیش­ترین محتوای پرولین، قندهای محلول و آنتوسیانین در بالاترین سطح شوری به­دست آمد. در بین سطوح پرایمینگ مورد مطالعه هیدروپرایمینگ در تنش شوری 200 میلی­مولار دارای بیش­ترین درصد و سرعت جوانه­زنی بذر، محتوای کلروفیل، آنتوسیانین و پرولین بود.

چکیده [English]

Salt tolerance at the germination stage is important, because salinity is usually severeat the soil surface.Especially in cases where the groundwater level is high.In order to investigate the effect of primingand salinity stress on the quantitative and qualitative traits of P. oleracea, this experiment was conducted in the Seed Technology Laboratory of the Faculty of Agricultural Sciences of Shahed University in 2019.Experimental agents included pretreatments (control (distilled water), hydro-priming, potassium nitrate and salicylic acid) in response to 6 salinity levels (no salinity as control and salinity of 30, 60, 90, 120 and 200 mM with Sodium Chloride).This experiment was studied as a factorial in a completely randomized design with three replications. The measured traits included germination indices, photosynthetic pigments, anthocyanin content, proline and soluble sugars.The results showed that the highest amount of germination components such as germination percentage (97%), germination rate (2.16 seeds per day), plantletlength (6.69 cm), plantletfresh weight (4 g), plantletlength index (324.69) and RWC (75.25%) were obtained in non-salinity stress and application of hydro-priming pretreatment.At salinity levels of 60 mM, the content of chlorophyll a, chlorophyll b and total chlorophyll in the hydropriming treatment increased and the highest content of proline, soluble sugars and anthocyanin was obtained at the highest salinity level.Among the studied priming levels, hydropriming at salinity stress of 200 mM had the highest percentage and seed germination rate, chlorophyll content, anthocyanin and proline.
