گروه‌بندی ژنوتیپ‌های لوبیاتحت تنش خشکی درمرحله جوانه‌زنی بذر و مطالعه بیان برخی از ژن‌ها در ژنوتیپ‌های حساس و متحمل

چکیده

به­منظور تعیین ژنوتیپ متحمل و حساس به خشکی لوبیا در مرحله جوانه‌زنی، آزمایشی به­صورت فاکتوریل دو عاملی در قالب طرح کاملا تصادفی با سه تکرار در سال 1394 اجرا شد. عامل اول شامل ژنوتیپ (21 ژنوتیپ)، عامل دوم تنش خشکی (با استفاده از پلی‌اتیلن­گلیکول 6000 در 2 سطح صفر و 148/0- مگاپاسکال) بودند. نتایج تجزیه واریانس برای صفات مورد بررسی (شامل طول ریشه‌چه، طول ساقه‌چه، وزن تر ریشه‌چه، وزن خشک ریشه‌چه، وزن تر ساقه‌چه، وزن خشک ساقه‌چه، سرعت رشد ساقه‌چه، سرعت رشد ریشه‌چه، نسبت طول ریشه‌چه به ساقه‌چه، اختلاف وزن خشک و تر ساقه‌چه و اختلاف وزن خشک و تر ریشه‌چه) نشان داد که اثر ژنوتیپ‌ و همچنین اثر متقابل ژنوتیپ و خشکی، بر کلیه صفات مورد بررسی معنی‌دار بودند. بر اساس نتایج حاصل از تجزیه خوشه‌ای به‌روش Ward، ژنوتیپ‌های مطالعه‌شده تحت شرایط نرمال و تنش خشکی از نظر خصوصیات گیاهچه­ای در چهار گروه مختلف قرار گرفتند که ژنوتیپ­های غلاف سیاه و پاک به‌ترتیب به‌عنوان ژنوتیپ­های متحمل و حساس در شرایط تنش خشکی انتخاب شدند. بررسی الگوی بیان سه ژن GR، PRX و APX واکنش‌دهنده به تنش خشکی در دو ژنوتیپ پاک و غلاف سیاه لوبیا با استفاده از تکنیک Real time PCR مورد مطالعه قرار گرفت. پس از استخراجRNA  و سنتز cDNA، تظاهر بیان سه ژن در تیمارهای صفر، 72، 144، 192 و 240 ساعت خشکی نشان داد که ژن­های واکنش‌دهنده به خشکی در این پژوهش بیان بالاتری در رقم متحمل نسبت به رقم حساس داشت و میزان بیان سه ژن PRX، GR و APX در ساعت انتهای تنش، در ژنوتیپ متحمل به‌ترتیب دو، چهار و شش برابر بیش‌تر از ژنوتیپ حساس بود.

چکیده [English]

In order to study the effects of drought stress induced by polyethylene glycol 6000 on germination components of 21 bean genotypes, a randomized complete design with factorial arrangement of one drought level (-0.148 MPa) and control level (0), in 3 replication and with the purpose of determining the resistance and susceptible genotype, was conducted. Results of anova indicate that all genotypes in all studied components (root, shoot, fresh weight of root and shoot, dry weight of root and shoot, growth rate root and shoot, root to shoot length ratio, difference of fresh and dry weight root, difference of fresh and dry weight shoot)were affected significantly by drought stress. Genotype effect and also the interaction between genotype and drought, was significant for all studied components. According to results of cluster analysis with the ward method, all genotypes in normal and drought conditions, were placed into four different groups, the Ghalaf Siyah and Pak genotypes were chosen as resistance and susceptible genotypes in drought stress conditions.In this study the expression patterns of three drought induced-genes was investigated in two Ghalaf Siyah (resistant genotype) and Pak (sensitive genotype) using Real time PCR technique. After RNA extraction and cDNA synthesis, expression of three genes, including,GR, PRX and APX were studied in the seedling stage of Ghalaf Siyah and Pak genotype under drought stress for control, 72, 144, 192 and 240 hours of the treatments.The results indicated that in Ghalaf Siyah genotype the drought  induced-genes generally had higher expression than Pak cultivar.Expression of three genes PRX, GR and APX at the 240 h in the tolerant genotype, it was two, four and six more than the sensitive genotype.
