بررسی تاثیر هالوپرایمینگ بر جوانه‌زنی و رشد گیاهچه ذرت دانه‌ای سینگل کراس 704 در شرایط تنش شوری

چکیده

این پژوهش به­منظور بررسی تاثیر پرایمینگ بذر بر جوانه­زنی و خصوصیات فیزیولوژیک گیاهچه ذرت در قالب دو آزمایش جداگانه به­صورت آزمایش فاکتوریل در قالب طرح آماری کاملاً تصادفی با چهار تکرار انجام شد. در هر دو آزمایش، عامل اول محلول پرایمینگ حاصل از نمک‌های سولفات سدیم، کلرید سدیم و نیترات پتاسیم به غلظت دو درصد و عامل دوم سطوح تیمار شوری (0، 40 و 80 میلی‌مولار کلرید سدیم) بود. نتایج نشان داد تنش شوری در بذرهای پرایم­شده و پرایم­نشده سبب کاهش درصد جوانه‌زنی و رشد گیاهچه، فعالیت آنزیم آلفا آمیلاز و پایداری غشا سلولی گیاهچه ذرت شد، البته پرایمینگ بذر با محلول­های نمکی سبب کاهش تاثیر منفی تنش شوری بر رشد گیاهچه ذرت شد. پرایمینگ بذر با محلول نیترات پتاسیم و کلرید سدیم در مقایسه با نمک سولفات سدیم تاثیر مثبتی بر درصد جوانه‌زنی، طول گیاهچه، فعالیت آنزیم‌های آنتی‌اکسیدانت، غلظت اسید جیبرلیک و اکسین و غلظت پتاسیم گیاهچه ذرت در شرایط تنش شوری داشت. در سطح تنش شوری80 میلی­مولار کلرید سدیم، بیش­ترین طول گیاهچه در بذرهای تیمارشده با نمک‌های نیترات پتاسیم به­میزان 13 سانتی‌متر مشاهده شد. در شرایط تنش شوری شدید، با افزایش فعالیت آنزیم‌های آنتی‌اکسیدانت، کم­ترین غلظت مالون­دی­آلدئید بافت گیاهچه در بذرهای تیمارشده با نیترات پتاسیم (022/0 نانومول بر گرم بافت تازه برگ) مشاهده شد. همچنین پرایمینگ بذر ذرت با محلول نیترات پتاسیم توانست از روند کاهش شدید غلظت اسید جیبرلیک و اکسین تحت تاثیر تنش شوری جلوگیری نماید. در مجموع بذرهای پرایم­شده با نیترات پتاسیم بیش­ترین رشد گیاهچه ذرت در شرایط تنش شوری را داشتند.

چکیده [English]

This study was carried out to investigate the effect of corn seed priming on germination and physiological characteristics of corn seedling (SC 704) in two separate experiments, each in a factorial experiment with completely randomized design with four replications. In both experiments, the first factor was the priming solution of Sodium Sulfate, Sodium Chloride and Potassium Ntrate at 2% concentration and the second factor was the salinity treatment (0, 40 and 80 mM Sodium Chloride). The results showed that salt stress decreased germination percentage and seedling growth, α-amylase activity and cell membrane stability of maize seeds in primed and non- primed seeds, whereas priming with Potassium Nitrate solution and Sodium Chloride had a positive effect on germination percentage, seedling length, antioxidants enzymes activity, Gibberellic acid and Auxin concentration and potassium concentration of maize seedling in stress conditions. At 80 mM Sodium Chloride level, the maximum seedling length was observed in seeds treated with Potassium Nitrate (13 cm). Under severe salinity conditions, with increasing activity of antioxidant enzymes, the lowest malondialdehyde concentration was observed in seeds treated with Potassium Nitrate (0.02 nmol / g fw). Also, priming of maize seed with Potassium Nitrate solution prevented the reduction of gibberellic acid and auxin concentration under salinity stress. In conclusion, seeds which are treated with Potassium Nitrate had the highest corn seedling growth under salinity stress.
