مقایسه تاثیر تکنیک‌های مختلف پرایمینگ بر بهبود جوانه‌زنی و فعالیت آنزیم‌های آنتی‌اکسیدانت در گیاهچه‌های گاوزبان اروپایی

چکیده

یکی از مشکلات تولید گیاهان دارویی از قبیل گاوزبان اروپایی، جوانه­زنی غیریکنواخت و استقرار ضعیف گیاهچه­ها است. پرایمینگ یکی از راهکارها جهت‌ بهبود جوانه‌زنی و رشد گیاهچه‌ها در گیاهان زراعی و دارویی است. این پژوهش به­منظور بررسی تاثیر پرایمینگ بر جوانه­زنی، رشد و خصوصیات بیوشیمیایی و آنزیمی گیاهچه­های گاوزبان اروپایی انجام گرفت. تیمارهای آزمایشی شامل هیدروپرایمینگ به­مدت 48 ساعت، ویتامین‌پرایمینگ (اسید‌آسکوربیک 85/0 میلی­مولار) به­مدت 48 ساعت، هورمون‌پرایمینگ (اسید‌سالسیلیک 4 میلی­مولار) به­مدت 60 ساعت، هالوپرایمینگ (نیترات پتاسیم 20 میلی­مولار) به­مدت 24 ساعت و شاهد بودند. نتایج نشان داد که کلیه تکنیک‌های پرایمینگ مورد استفاده، باعث افزایش معنی‌دار جوانه‌زنی، رشد، شاخص‌های بیوشیمیایی و آنزیمی گیاهچه‌ها گردید. همچنین استفاده از روش‌های مختلف پرایمینگ در بذرهای گاوزبان اروپایی، موجب کاهش معنی‌‌دار میانگین زمان‌جوانه‌زنی و درصد گیاهچه‌های غیرنرمال گردید. بیش­ترین درصد و سرعت جوانه­زنی، شاخص قدرت و طول گیاهچه‌ از بذرهای هیدروپرایم­شده به­دست آمد که به­طور معنی­داری بیش­تر از بذرهای شاهد بودند. همچنین، هیدروپرایمینگ موجب کاهش 80 درصدی گیاهچه­های غیرنرمال و 25 درصدی میانگین ‌زمان‌جوانه‌زنی گردید. فعالیت آنزیم‌های کاتالاز، پراکسیداز و میزان اسیدآمینه پرولین در گیاهچه‌های حاصل از بذرهای پرایم‌شده به‌طور معنی‌داری بیش­تر از شاهد بود. بیش­ترین فعالیت آنزیم کاتالاز و محتوی پرولین به­ترتیب در ‌تیمار بذر با نیترات پتاسیم 20 میلی­مولار و اسید ‌سالسیلیک 4 میلی­مولار مشاهده شد که اختلاف معنی‌داری با هیدروپرایمینگ نداشتند. هیدروپرایمینگ موجب افزایش 7/2 برابری فعالیت آنزیم‌ پراکسیداز گیاهچه‌ها گردید. به­طور کلی، در بین ‌تیمار‌ها، هیدروپرایمینگ به­مدت 48 ساعت را می­توان به‌عنوان بهترین تیمار برای بهبود خصوصیات جوانه‌زنی، رشد، خصوصیات بیوشیمیایی و آنزیمی گاوزبان اروپایی در نظر گرفت.

چکیده [English]

One of the problems of producing medicinal plants, such as borage, is unequal and poor germination and seedling establishment. Priming is one of the technique for improving seed germination and seedling growth in crops and medicinal plants. This research was carried out in order to study the effect of seed priming on germination, growth and biochemical and enzymatic characteristics of borage seedlings. The Experimental treatments were hydro-priming for 48 hours, seed priming with 0.85 mM ascorbic acid for 48 hours, 4 mM salicylic acid concentration for 60 hours, 20 mM potassium nitrate (KNO3) concentration for 24 hours and control. The results showed that seed priming were significantly increased the seed germination, growth and biochemical characteristic of borage seedlings. The mean germination time and the percentage of abnormal seedlings were significantly decreased by priming the borage seeds. The highest percentage and rate of germination, vigor index and length of seedlings were obtained from hydro-priming seeds, which were significantly higher than control. Also, hydro-priming decreased the abnormal seedlings about caused 80% and mean germination time about 25%. The activity of catalase and peroxidase enzymes and proline in seedlings from primed seeds were significantly higher than those of the control. The highest activity of catalase enzyme and proline content were observed in seed priming of 20 mM KNO3 concentration for 24 hours and 4 mM salicylic acid for 60 hours respectively, which did not have significant difference with hydro-priming. Hydro-priming, caused increase 2.7 fold in the activity of peroxidase enzymes seedlings. Generally, among the treatments, hydro-priming for 48 hours for improving germination, growth and biochemical characteristics of borage is considered as the best treatment.
