تأثیر اسید سالیسیلیک‌ بر شاخص‌های جوانه‌زنی، بنیه بذر و فعالیت برخی آنزیم‌های آنتی‌اکسیدان آفتابگردان (Helianthus annuus L.) رقم آرماویرسکی تحت تنش خشکی

چکیده

از آن­جایی­که تولید گیاهان روغنی می‌تواند تحت تأثیر عوامل محیطی مانند محدودیت آب قرار گیرد، لذا تیمار با اسید سالیسیلیک‌ به‌عنوان یک تنظیم­کننده ‌رشد، قادر است مقاومت به خشکی را در گیاهان افزایش دهد. در این راستا، به‌منظور بررسی تأثیر اسید سالیسیلیک‌ بر برخی خصوصیات بیوشیمیایی و جوانه‌زنی بذر گیاه آفتابگردان در شرایط تنش خشکی، آزمایشی به‌صورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار در آزمایشگاه فیزیولوژی دانشگاه آزاد اسلامی واحد کلیبر در سال زراعی 1396 اجرا گردید. تیمارهای آزمایش شامل غلظت‌های مختلف اسید سالیسیلیک‌ در چهار سطح (صفر، 5/0، 1 و 5/1 میلی‌مولار) و سطوح مختلف تنش خشکی ایجادشده با پلی­اتیلن گلایکول6000 در پنج سطح با پتانسیل‌های صفر (آب مقطر)، 5-، 10-، 15- و 20- مگاپاسکال) بودند. نتایج نشان داد که اثر تنش خشکی و غلظت‌های مختلف اسید سالیسیلیک‌ بر مؤلفه‌های جوانه­زنی و آنزیم‌های آنتی‌اکسیدانت در سطح احتمال یک درصد معنی‌دار بود. برهمکنش تنش و اسید سالیسیلیک‌ بر میزان فعالیت آنزیم‌های سوپراکسید دیسموتاز، آسکوربات پراکسیداز، بنیه بذر، طول ریشه‌چه و ساقه‌چه معنی‌دار بود. نتایج مقایسه میانگین داده‌ها نشان داد که با افزایش تنش خشکی درصد و سرعت جوانه‌زنی کاهش و میزان فعالیت آنزیم‌های آنتی‌اکسیدانت افزایش یافت. در پژوهش انجام­شده کاربرد پیش­تیمار اسید سالیسیلیک‌ با غلظت‌های 1 و5/0 میلی‌مولار باعث افزایش معنی‌دار فعالیت آنزیم‌های مالون ‌دی‌آلدئید، سوپراکسید دیسموتاز، گلوتاتیون پراکسیداز، کاتالاز، آسکوربات پراکسیداز و مؤلفه‌های جوانه­زنی در مقایسه با شاهد گردید. نتایج کلی این آزمایش نشان داد که محلول‌پاشی اسید سالیسیلیک‌ به‌ویژه در غلظت‌ 1 میلی‌مولار با کاهش تنش اکسیداتیو ناشی از خشکی سبب بهبود شاخص‌های جوانه‌رنی و افزایش در فعالیت آنزیم‌های آنتی‌اکسیدانت در بذر آفتابگردان شد.

Since the production of oily plants can be affected by environmental factors such as water restriction, so treatment with acid salicylic as a growth regulator can increase drought tolerance in plants. In order to investigate the effect of acid salicylic on some biochemical and seed germination of sunflower seeds under drought stress conditions, an experiment was conducted as factorial in a completely randomized design with three replications in the physiology lab at the Islamic Azad University of Kaleiber Branch in 2017.The treatments consisted of different concentrations of acid salicylic in four levels (0, 0.5, 1 and 1.5 mM) and different levels of drought stress with polyethylene glycol 6000 at five levels with Zero potential of (distilled water), -5, -10, -15 and -20 megapascal). The results showed that the effect of drought stress and different concentrations of acid salicylic on germination and antioxidant enzymes was significant at 1% probability level. Interaction of stress and acid salicylic on the activity of superoxide dismutase enzymes, ascorbate peroxidase, seed vigor, root length and stem length were significant. In drought stress conditions, the plant has been shown to increase the activity of antioxidant enzymes to reduce the effects of oxidative stress. The results of the mean comparison of the data showed that with increasing drought stress, the percent and speed of germination decreased and the activity level of antioxidant enzymes increased. In the present study, acid salicylic pre-treatment with concentrations of 1 and 0.5 mM significantly increased the activity of malondialdehyde, superoxide dismutase, glutathione peroxidase, catalase, ascorbate peroxidase enzymes and germination components in comparison with the control.The overall results of this experiment showed that acid salicylic spraying, especially at 1 mM concentration, reduced the oxidative stress caused by drought and improved the antioxidant enzymes activity in sunflower seeds.
