ارزیابی مولفه‌های جوانه‌زنی، آنزیم‌های آنتی‌اکسیدانت و هدایت الکتریکی بذرهای لوبیا چیتی (Phaseolus vulgaris L) بدست آمده از پیش‌تیمار بذرهای مختلف گیاه مادری

چکیده

به­دلیل نقش جوانه­زنی بذر در استقرار بوته، جوانه­زنی مطلوب به­عنوان یک عامل کلیدی در کشاورزی اهمیت ویژه­ای دارد. به­منظور بررسی مولفه­های جوانه­زنی، آنزیم­های آنتی­اکسیدانت و هدایت الکتریکی بذرهای بدست آمده از پیش­تیمار بذر گیاه مادری آزمایشی به­صورت فاکتوریل در قالب طرح بلوک کاملا تصادفی با 3 تکرار در سال 1393 در دانشگاه زابل انجام گرفت. تیمار­های آزمایش شامل عامل رقم لوبیای چیتی در 3 سطح E9، E10 و خمین و عامل دوم بذرهای به­دست آمده از پیش­تیمار بذر گیاه مادری در مزرعه در 7 سطح 1- پلی­اتیلن­گلیکول 5 -بار در مدت 6 ساعت 2-کلرید پتاسیم 20– میلی­مولار در 6 ساعت 3- کلرید کلسیم 15- میلی­مولار در 3 ساعت 4- کلرید سدیم 20- میلی­مولار در 6 ساعت 5 – اسید سالیسیلیک 500 میلی­گرم در لیتر در 12 ساعت 6-آب خالص در مدت 12 ساعت و 7- بدون پیش­تیمار (شاهد) بود. صفات درصد و سرعت جوانه­زنی، شاخص وزنی و طولی بنیه گیاهچه، انرژی جوانه­زنی در روز چهارم، آلفا آمیلاز، سوپر اکسید دیسموتاز، قند محلول، پراکسیداز و هدایت الکتریکی اندازه­گیری شد. نتایج نشان داد که اثر متقابل پیش­تیمار و رقم بر صفات درصد جوانه­زنی، شاخص وزنی بنیه و انرژی جوانه­زنی روز چهارم در سطح 1% و بر شاخص طولی بنیه در سطح 5% معنی­دار بود. همچنین صفات آلفاآمیلاز، سوپر اکسید دیسموتاز، قند محلول، پراکسیداز و هدایت الکتریکی در سطح 1% معنی­دار بود. بیش­ترین درصد و سرعت جوانه­زنی در تیمار کلرید کلسیم 3 ساعت رقم E9، بیش­ترین شاخص بنیه در تیمار کلرید کلسیم 3 ساعت رقم E10 و قند محلول، پراکسیداز و آلفا آمیلاز از تیمار کلرید کلسیم 3 ساعت به­دست آمد. بذر­های حاصل از پیش­تیمار گیاه مادری از قدرت جوانه­زنی خوبی برخوردارند و با توجه به رقم و انتخاب پیش­تیمار مناسب بذر­های گیاه مادری، می­توانیم جوانه­زنی و استقرار و رشد گیاهچه لوبیا را بهبود بخشیم.

Due to the role seeds germination of the plant establishment, optimal germination as a key factor in agriculture is important. In order to determine the components of germination, enzymes, antioxidants and electrical conductivity seeds obtained from pretreatment a mother plant seed on the farm a factorial experiment in a completely randomized design with three replications was conducted in the laboratory of Bio-center, College of Agriculture, Zabol University. first factor include varieties at 3 levels (Pinto bean varieties E9, E10, and Khomain) and second factor seeds obtained from pretreatment the mother plant seed on the farm at 7 levels: 1-(polyethylene glycol (PEG) -5 bar at 6 hours, 2- salicylic acid (C7H6O3) 500 mg per liter at 12 hours, 3- potassium chloride (Kcl) -20 mM at 6 hours, 4-calcium chloride( Cacl2) - 15 mM at 3 hours, 5- sodium chloride (Nacl) -20 mM at 6 hours, 6-  distilled water at 12 hours and 7- without pretreatment (control). Percentage and rate germination, weight and lengh vigor index, energy germination 4th days, Alpha-amylase, Sugar solution, Peroxidase, Peroxidase and super oxide despotize were measured. The result showed that the interaction between pretreatment and varieties on germination traits include germination percentage, weighting vigor index, Energy Germination 4th days at 1% and vigor index length at 5% were significant. The biochemical characteristics of seeds obtained from pretreatment the mother plant seed on the farm was significant at 1%. The highest percentage and germination rate were obtained calcium chloride at 3 hours varieties E9, highest vigor index on calcium chloride at 3 hours varieties E10 and soluble sugar, alpha-amylase, peroxidase on calcium chloride at 3 hours.  The result showed pretreatment Cacl2 at 3 hours improved the germination and growth seedling on bean. Pretreatment seeds of mother plant are good germination and according to variety and selection of appropriate pretreatment for mother plant, can improve germination, establishment and growth seedling beans
