Evaluation of different hormonal and wet chilling time treatments on biochemical and germination indices in seeds of three species of Artemisia (A.annua, A.biensis willd., A.vulgaris L.)

Abstract

Artemisia sp. from Asteraceae family is one of the important medicinal and pasture plants that have many uses in various medicinal and food industrial. Seeds of this plant have dormancy so this study was performed to evaluate the effects of different hormonal treatments on breaking dormancy in three species of annual (A.annua), biennial (A.biensis) and perennial (A.vulgare) of Artemisia under different wet chilling times. This research was conducted in a split plot layout experiment in a completely randomized design (CRD) with 3 replications and 2 separate repetition in Herbal Research Institute, Islamic Azad University Branch of Shahrekord in 2024. Experimental treatments included wet chilling (A1-A4: 0, 10, 20, 30 day) in main plots and hormones inclusive (gibberellic acid (GA3) (120ppm), benzyl adenine (BA) (10-4 M), kinetin (KIN) (10-4 M), GA3 (120ppm)+ BA (10-4 M), GA3 (120ppm)+KIN (10-4 M), BA (10-4 M)+KIN (10-4 M), GA3 (120ppm)+BA (10-4 M)+KIN (10-4 M) and control) in sub-plots. Biochemical indices such as hydrogen peroxide, alpha-amylase and beta-1, 3 glucanase and germination indices such as dry weight of seedlings, length of seedlings, germination percentage, mean of germination time, seed vigour and germination index were measured. According to the obtained results, utilization of wet chilling treatment, significantly increased the germination characters and decrease the mean of germination time. The most and lowest important measured characters such as Seedling length (11.12-4.19 cm), seed vigor (7.9-1.46), seedling dry weight (101.14-39.88 mg), germination percentage (71.12-29.63), germination index (18.66-3.25), content of alpha-amylase (11.77-2.41 mM/gr fresh weight of seed), beta-1, 3 glucanase (9.81-2.57 mM/gr fresh weight of seed) and hydrogen peroxide (0.63-0.191 mM/gr fresh weight of seed) obtained in wet chilling in 20 and 30 day+GA3 (120ppm)+BA (10-4M)+ KIN (10-4M) and treatment of without wet chilling priming+without hormonal, respectively, so that the amounts of these characters in this treatments more than 100% were more than the control seeds. Results showed that in most of measured characters the A.annua better than A.biensis and the specie of A.biensis better than A.vulgaris. The most of germination percentage and other characters made in A.annua and other wise the A.vulgaris had the lowest measured characters. Generally wet chilling (20 day) with use of GA3, BA and KIN suggest to best seed germination of Artemisia sp.
