تأثیر پرایمینگ و فرسودگی بذر بر خصوصیات جوانهزنی، ویژگیهای بیوشیمیایی و بیان ژنهای آنزیمهای آنتیاکسیدانت در لوبیا Phaseolus Vulgaris (L.)
بهمنظور بررسی اثر پرایمینگ و فرسودگی (پیری تسریعشده) بذر بر جوانهزنی و ترکیبات بیوشیمیایی گیاه لوبیا آزمایشی بهصورت فاکتوریل در قالب طرح کاملاً تصادفی در دانشگاه محقق اردبیلی در سال 1395 با 3 تکرار انجام شد. تیمارها شامل فرسودگی (شاهد و دو سطح 88 و 78 درصد جوانهزنی) و پرایمینگ (بدون پرایمینگ، هیدرو پرایمینگ، پرایمینگ با جیبرلین و اسید سالیسیلیک) بود. پس از استخراج RNA و ساخت cDNA بررسی بیان ژنهای آنزیمهای آنتیاکسیدانت با استفاده از qRT-PCR، و برخی شاخصهای فیزیولوژیک و بیوشیمیایی سنجیده شد. نتایج نشان داد که بیشترین طول ریشهچه و ساقهچه، وزنتر ریشهچه و ساقهچه، وزن خشک ریشهچه و ساقهچه، میزان ذخایر فسفات از تیمار پرایمینگ با جیبرلین و بدون فرسودگی حاصل شد. همچنین بیان ژن آنزیم پراکسیداز در تیمار پرایمینگ با جیبرلین و سطح فرسودگی 88 درصد بالاتر از سایر آنزیمهای آنتیاکسیدانت بود. با افزایش فرسودگی بیان ژن کاهش پیدا کرد بهطوریکه کمترین میزان بیان ژن آنزیم پراکسیداز در تیمار بدون پرایمینگ و سطح فرسودگی 78 درصد بود. در نتیجه، استفاده از پیشتیمار جیبرلین موجب تقویت بذرهای ضعیف لوبیا میشود.

اثر آللوپاتیک گیاهان بر برخی شاخصهای جوانهزنی، فعالیت بیوشیمیایی و آنزیمی علفهای هرز
بهمنظور بررسی اثر آللوپاتی عصاره آبی گیاه سورگوم، اکالیپتوس، آفتابگردان و تنباکو بر برخی شاخصهای جوانهزنی، بیوشیمیایی و آنزیمی دو علف هرز یولاف وحشی و خردل وحشی، آزمایشی بهصورت فاکتوریل در قالب طرح پایه کاملاً تصادفی با 3 تکرار، در سال 1396 در آزمایشگاه تکنولوژی بذر دانشکده کشاورزی دانشگاه شاهد، اجرا شد. چهار غلظت (شاهد، 5، 10 و 20 درصد) از چهار گیاه (سورگوم، اکالیپتوس، آفتابگردان و تنباکو) و دو نمونه بذر علف هرز (خردل وحشی و یولاف وحشی) تیمارهای آزمایش بودند. نتایج تجزیه واریانس دادهها نشان داد که درصد و سرعت جوانهزنی، طول گیاهچه، وزن خشک گیاهچه، پراکسیداسیون لیپید و فعالیت کاتالاز دو علف هرز در سطح آماری یک درصد تحت تأثیر غلظتهای متفاوت عصاره آبی گیاهان قرار گرفت. نتایج نشان داد با افزایش غلظت عصاره آبی گیاه میزان درصد و سرعت جوانهزنی، طول گیاهچه، وزن خشک گیاهچه، فعالیت آنزیم کاتالاز هر دو علف هرزکاهش یافت، درحالیکه میزان پراکسیداسیون لیپید آنها زیاد شد. در غلظتهای متفاوت عصاره آبی گیاه، علف هرز یولاف وحشی نسبت به خردل وحشی بیشترین میزان تأثیرپذیری را داشت و بیشترین تأثیر بازدارندگی در غلظت 20 درصد مشاهده شد. غلظتهای متفاوت عصاره آبی سورگوم نسبت به دیگر عصاره آبی گیاهان اثر مهاری بیشتری بر هر دو علف هرز داشتند بهطوریکه دیده شد غلظت 20 درصد منجر به مهار بیشتر هردو علف هرز شد و این عصاره آبی بیشترین اثر آللوپاتیک را نشان داد که این امر میتواند نویدی برای تولید علفکشهای با منشاء طبیعی باشد.

بهمنظور بررسی تأثیر هیومیک اسید و فولویک اسید بر خصوصیات فیزیولوژیک گیاهچه سرخارگلتحت شوری، آزمایشی بهصورت فاکتوریل در قالب طرح پایه کاملاً تصادفی با سه تکرار در آزمایشگاه تکنولوژی بذر دانشگاه شاهد در سال 1395 اجرا شد. تیمارهای آزمایش شامل تنش شوری در چهار سطح (0، 4، 8 و 12 دسیزیمنس بر متر)، هیومیک اسید در سه سطح (0، 200 و 400 میلیگرم بر لیتر) و فولویک اسید در سه سطح (0، 200 و 400 میلیگرم بر لیتر) بود. صفات مورد بررسی شامل درصد جوانهزنی، محتوای نسبی آب، محتوای کلروفیل a، b، کل و محتوای کارتنوئید، پرولین و پروتئین بود. با توجه به نتایج تجزیه واریانس، اثر متقابل هیومیک اسید، فولویک اسید و تنش شوری بر صفات مورد بررسی به استثنای محتوای نسبی آب گیاهچه معنیدار بود. کاربرد هیومیک اسید 200 میلیگرم بر لیتر و فولویک اسید 400 میلیگرم بر لیتر با میانگین 55/75 درصد بالاترین درصد جوانهزنی را در تنش شوری ds/m 12 داشت. بر اساس نتایج تحقیق حاضر، هیومیک اسید بیشترین تأثیر را بر محتوای ترکیبات محلول مورد مطالعه داشت، بهگونهای که تا زمانی غلظت فولویک اسید 200 میلیگرم بر لیتر یا صفر بود میتوان افزایش ترکیبات محلول را در سطوح هیومیک اسید مشاهده کرد. غلظت 400 میلیگرم بر لیتر هیومیک اسید در عدم حضور فولویک اسید و تنش ds/m 12 دارای بیشترین محتوای کلروفیل کل بود. تنش شوری باعث کاهش جوانهزنی بذر سرخارگل شد، کاربرد هیومیک اسید و فولویک اسید اثرات ناشی از تنش شوری را، هر چند اندک بهبود بخشیدند.

اثر امواج فراصوتی بر جوانهزنی، فعالیت برخی آنزیمها و ویژگیهای بیوشیمیایی بذر سرخارگل
یکی از روشهای بیوفیزیکی پرایمینگ بذر، پیشتیمار بذرها با امواج فراصوتی میباشد. ضربههای ممتد امواج فراصوتی سبب افزایش نفوذپذیری پوسته بذر، تسریع جذب آب و بالارفتن دمای بافتها میشود. بالارفتن سرعت آماس و دمای بافتهای بذرهای تیمارشده با امواج فراصوتی احتمالا با تسریع در تغییرات متابولیسم بذر همراه میباشد. جوانهزنی بذور سرخارگل ضعیف و نامنظم میباشد که ناشی از خواب بذر میباشد. بنابراین بهمنظور بررسی اثر امواج فراصوتی بر جوانهزنی، فعالیت برخی آنزیمها و پارامترهای بیوشیمیایی جوانهزنی بذر سرخارگل، آزمایشی در سال 1396 در آزمایشگاه زراعت دانشگاه صنعتی شاهرود در قالب طرح پایه کاملاً تصادفی با 5 تیمار و 4 تکرار انجام شد. تیمارهای آزمایشی شامل 5 مدت تیمار امواج فراصوتی 0، 10، 15، 20 و 25 دقیقه با دستگاه التراسونیک با قدرت ثابت 50 وات و دمای 25 درجه سلسیوس بود. نتایج تجزیه واریانس دادهها نشان داد که امواج فراصوتی بر صفات اندازهگیریشده اثر معنیداری در سطح احتمال یک درصد نشان داد. امواج فراصوتی فعالیت آنزیمهای آلفاآمیلاز، پروتئاز، الکلدهیدروژناز، گلوکز 6- فسفاتدهیدروژناز، غلظت پروتئین کل، محلول و قند کل را در اغلب تیمارها نسبت به شاهد افزایش داد. با افزایش فعالیت آنزیمهای آلفاآمیلاز، پروتئاز، الکلدهیدروژناز، آرژیناز و گلوکز 6- فسفات دهیدروژناز درصد جوانهزنی، سرعت جوانهزنی و وزن خشک گیاهچه افزایش یافت. در بیشتر صفات اندازهگیریشده مدت 15 دقیقه تیمار با امواج فراصوتی بیشترین میزان فعالیت آنزیمها و شاخصهای جوانهزنی را نشان داد، بنابراین مدت 15 دقیقه تیمار امواج فراصوتی در دمای 25 درجه سلسیوس و قدرت 50 وات برای پیشتیمار بذر سرخارگل پیشنهاد میشود.

بهمنظور مطالعه اثر تنش شوری بر روی برخی خصوصیات بیوشیمیایی و مورفوفیزیولوژیک ارقام مختلف گندم در مرحله گیاهچهای، آزمایشی بهصورت فاکتوریل در قالب طرح پایه کاملاً تصادفی با سه تکرار در آزمایشگاه تحقیقات دانشکده کشاورزی دانشگاه زابل در سال 1395 اجرا گردید. تیمارهای آزمایش شامل تنش شوری در چهار سطح شاهد و 100، 200 و 300 میلیمولار کلریدسدیم و 11 رقم مختلف گندم نان و یک رقم گندم دوروم بودند. ارقام از نظر تمامی صفات بجز هدایت روزنهای دارای اختلاف معنیدار در سطح یک درصد بودند. همبستگی مثبت و معنیداری بین درصد جوانهزنی و سرعت جوانهزنی و همبستگی منفی و معنیداری بین فعالیت کاتالاز و مقدار کربوهیدرات در شرایط تنش شوری مشاهده شد. تجزیه به عاملها بر اساس تجزیه به مؤلفههای اصلی و چرخش وریماکس در شرایط بدون تنش شوری و در شرایط تنش شوری (متوسط تنشها) هرکدام پنج عامل بهترتیب 13/87 و 11/83 درصد از تغییرات را تبیین کردند. نتایج حاصل از تجزیه ضرایب عامل نشانگر اهمیت مؤلفه تحمل به تنش شامل مقدار پرولین، فعالیت آنزیمهای آسکوربات پراکسیداز و گلوتاتیون پراکسیداز، طول و تعداد ریشهچه در گزینش ارقام مطلوب برای شرایط تنش شوری بود. ارقام مورد نظر با استفاده از تجزیه خوشهای بهروش وارد در هر دو شرایط عدم تنش و تنش شوری، دو گروه را تشکیل دادند. با توجه به نتایج، میتوان از تلاقی ارقام گروه اول با گروه دوم بهعلت حداکثر اختلاف، جهت ایجاد تنوع ژنتیکی استفاده نمود.

ردیابی Xanthomonas axonopodis pv. phaseoli و استفاده از روش بهینهشده در پایش سلامت تودههای بذری لوبیا
بیماری سوختگی باکتریایی معمولی لوبیا با عاملXanthomonas axonopodis pv. phaseoli از بیماریهای مهم بذرزاد لوبیا در اکثر مناطق کشت دنیا محسوب میشود. استفاده از بــذر سالم و گواهیشده از راهکارهای اصلی مقابله با گسترش این بیماری به شمار میرود، لذا ارزیابی سلامت بذور در بسیاری از کشورها بهصورت روتین در برنامههای گواهی انجام میشود. در تحقیق حاضر آزمونهای تشخیصی کشت روی محیط انتخابی، PCR و بیماریزایی بر روی رقم حساس خمین برای ردیابی باکتریXanthomonas axonopodis pv. phaseoli بهینهسازی شد. این روشهای بهینهشده برای بررسی سلامت پارتهای بذری لوبیا استفاده شدند. نتایج نشان داد که استفاده از روش کشت روی محیط انتخابی پیش از آزمون PCR، بهمیزان زیادی خطای تمایز کلنیهای مشابه غیر هدف از کلنیهای بیماریزای باکتری را کاهش میدهد. همچنین انجام آزمون PCR روی کلنیهای مشکوک جداشده از محیطهای انتخابی در مقایسه با PCR بر روی عصاره بذر بهدلیل حذف ممانعتکنندههای گیاهی بهمیزان بسیار زیادی، امکان واکنش کاذب دروغین را کاهش میدهد. روشهای مذکور برای بررسی سلامت نمونههای بذری منتخب از مزارع بذری استاندارد با موفقیت استفاده شد و نتایج حاکی از احراز سلامت بذور و عدم آلودگی به باکتریX. axonopodis pv. phaseoli در تودههای بذری منتخب گواهیشده کشور است.

تأثیر اسید سالیسیلیک بر شاخصهای جوانهزنی، بنیه بذر و فعالیت برخی آنزیمهای آنتیاکسیدان آفتابگردان (Helianthus annuus L.) رقم آرماویرسکی تحت تنش خشکی
از آنجاییکه تولید گیاهان روغنی میتواند تحت تأثیر عوامل محیطی مانند محدودیت آب قرار گیرد، لذا تیمار با اسید سالیسیلیک بهعنوان یک تنظیمکننده رشد، قادر است مقاومت به خشکی را در گیاهان افزایش دهد. در این راستا، بهمنظور بررسی تأثیر اسید سالیسیلیک بر برخی خصوصیات بیوشیمیایی و جوانهزنی بذر گیاه آفتابگردان در شرایط تنش خشکی، آزمایشی بهصورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار در آزمایشگاه فیزیولوژی دانشگاه آزاد اسلامی واحد کلیبر در سال زراعی 1396 اجرا گردید. تیمارهای آزمایش شامل غلظتهای مختلف اسید سالیسیلیک در چهار سطح (صفر، 5/0، 1 و 5/1 میلیمولار) و سطوح مختلف تنش خشکی ایجادشده با پلیاتیلن گلایکول6000 در پنج سطح با پتانسیلهای صفر (آب مقطر)، 5-، 10-، 15- و 20- مگاپاسکال) بودند. نتایج نشان داد که اثر تنش خشکی و غلظتهای مختلف اسید سالیسیلیک بر مؤلفههای جوانهزنی و آنزیمهای آنتیاکسیدانت در سطح احتمال یک درصد معنیدار بود. برهمکنش تنش و اسید سالیسیلیک بر میزان فعالیت آنزیمهای سوپراکسید دیسموتاز، آسکوربات پراکسیداز، بنیه بذر، طول ریشهچه و ساقهچه معنیدار بود. نتایج مقایسه میانگین دادهها نشان داد که با افزایش تنش خشکی درصد و سرعت جوانهزنی کاهش و میزان فعالیت آنزیمهای آنتیاکسیدانت افزایش یافت. در پژوهش انجامشده کاربرد پیشتیمار اسید سالیسیلیک با غلظتهای 1 و5/0 میلیمولار باعث افزایش معنیدار فعالیت آنزیمهای مالون دیآلدئید، سوپراکسید دیسموتاز، گلوتاتیون پراکسیداز، کاتالاز، آسکوربات پراکسیداز و مؤلفههای جوانهزنی در مقایسه با شاهد گردید. نتایج کلی این آزمایش نشان داد که محلولپاشی اسید سالیسیلیک بهویژه در غلظت 1 میلیمولار با کاهش تنش اکسیداتیو ناشی از خشکی سبب بهبود شاخصهای جوانهرنی و افزایش در فعالیت آنزیمهای آنتیاکسیدانت در بذر آفتابگردان شد.

اثر اسید هومیک، ورمیکمپوست و قارچ مایکوریزا بر برخی ویژگیهای مورفولوژیک و بیوشیمیایی گیاه دارویی ماریتیغال تحت تنش شوری
گیاه ماریتیغال یا خارمریم از گیاهان دارویی و یکسالهای است که در نقاط مختلف دنیا رشد میکند. این گیاه در درمان بیماریهای کبدی و جلوگیری از هپاتیت مورد استفاده قرار میگیرد. هدف این پژوهش بررسی اثر شوری، اسید هیومیک، ورمیکمپوست و دو نوع قارچ مایکوریزا بر ویژگیهای گیاه ماریتیغال است. این پژوهش در دو بخش جداگانه بهصورت بلوک کامل تصادفی با سه تکرار انجام شده است. برای انجام این پژوهش در بخش اول از دو گونه قارچ مایکوریزا شاملGlomus mosseae و Glomus intraradices استفاده شد و شوری در سه سطح 2، 4 و 8 dS/m به تیمارها اعمال شد. همچنین در بخش دوم از چهار سطح اسید هیومیک و سه سطح ورمیکمپوست و قارچ استفاده شده است. برخی از ویژگیهای مرفولوژیک و چهار آنزیم سوپراکسیددیسموتاز، پراکسیداز، کاتالاز و سلیبین اندازهگیری شد. نتایج حاصل از این پژوهش نشان داده است که با افزایش شوری ویژگیهای گیاه شامل طول ریشهچه، وزن خشک گیاهچه، درصد جوانهزنی، طول ساقهچه کاهش معنیداری یافته است و مدت زمان لازم برای جوانهزنی افزایش یافته است. همچنین بیشترین درصد جوانهزنی بذرها تیمار قارچ مایکوریزا Glomus intraradices در شرایط شوری با غلظت dS/m ۲ بهمقدار ۴۵/۹۸ درصد بوده است و کمترین مقدار درصد جوانهزنی نیز مربوط به تیمار قارچ مایکوریزا Glomus mossea در شرایط تنش شوری dS/m ۸ به مقدار ۳۲/۷۰ درصد بوده است. بالاترین فعالیت آنزیمها در تیمار 75 درصد ورمیکمپوست و 75 میلیگرم بر لیتر اسید هیومیک در شرایط رشد قارچ Glomus intraradices بهدست آمده است. استفاده از قارچ مایکوریزا Glomus mosseae و Glomus intraradices، ورمیکمپوست و اسید هیومیک سبب بهبود ویژگیها (ویژگیهای مورفولوژیک شامل طول ریشهچه، درصد جوانهزنی، وزن خشک و طول ساقهچه و ویژگیهای بیوشیمیایی شامل آنزیم سوپراکسید دیسموتاز، پراکسیداز، کاتالاز و سلیبین) در گیاه ماریتیغال شده است.

اثر دگرآسیبی خارشتر (L. Alhagi m`ourorum) بر خصوصیات جوانهزنی گندم (Triticum aestivum L.) و گلرنگ ((Carthamus tinctorius L.
این تحقیق با هدف بررسی اثر آللوپاتیک عصاری آبی و هیدروالکلی اندام هوایی گیاه دارویی خارشتر بر جوانهزنی و خصوصیات رشدی و محتوای پروتئین و قند محلول گلرنگ و گندم، بهصورت آزمایش فاکتوریل در قالب طرح کاملا تصادفی انجام شد. تیمارهای آزمایش شامل غلظتهای (0، 25/0، 50/0، 75/0 و 1 درصد) عصارهی اندام هوایی بودند. نتایج حاصل از تجزیه واریانس دادهها نشان داد که اثر غلظت عصارهها، نوع گیاه و غلظت عصاره× نوع گیاه در مورد صفات اندازهگیریشده معنیدار بود. بر اساس نتایج، استفاده از عصارهی آبی و هیدروالکلی اندام هوایی خارشتر باعث کاهش معنیدار درصد جوانهزنی، طول و وزن خشک ساقهچه و ریشهچه، مقدار پروتئین محلول و قند محلول در گلرنگ و گندم شد، با این حال عصارهی هیدروالکلی اثر بازدارندگی بیشتری بر گیاهان مورد آزمایش داشت. در مورد درصد جوانهزنی عصاره هیدروالکلی در گندم 30 درصد و در گلرنگ 66/33 درصد اثر کاهشی بیشتری نسبت به عصاره آبی داشت. با توجه به نتایج حاضر، بهنظر میرسد خارشتر بهعنوان علف هرز مزارع گندم و گلرنگ رشد و کیفیت محصول را بهطور معنیداری (05/0 >P ) کاهش میدهد.

بررسی نوع قارچکش و دوره انبارداری بر بنیه بذر سه رقم جو درطبقه مادری
بهمنظور تأثیر تیمارهای ضدعفونی بذر برقابلیت انبارمانی و شاخصهای بنیه بذر ارقام جو در طبقه مادری آزمایشی بهصورت فاکتوریل در قالب طرح پایه کاملاً تصادفی و در شرایط مزرعه بهصورت فاکتوریل با طرح پایه بلوک کامل تصادفی اجرا گردید. فاکتور اول شامل نوع قارچکش در سه سطح کاربوکسین تیرام با غلظت 5/2 در هزار، دیفنوکونازول+ سایپروکونازول (دیویدند استار) با غلظت 2 در هزار و شاهد (بدون ضدعفونی)، فاکتور دوم شامل سه رقم یوسف، نصرت و گوهران در طبقه مادری بودهاند. نتایج حاکی از آن است که درصد گیاهچههای عادی بعد از 2و 14 ماه انبارداری تحت تأثیر رقم در سطح احتمال 1 درصد قرار گرفت. بهطوریکه در این دو دوره بیشترین درصد گیاهچههای عادی در رقم نصرت و کمترین در رقم گوهران مشاهده گردید. بهعلاوه درصد گیاهچههای عادی بعد از 2ماه انبارداری تحت تأثیر تیمار ضدعفونی بذر در سطح احتمال 5 درصد معنیدار گردید. در مقایسه بین تیمارهای قارچکش از نظر تأثیر بر این صفت حداکثر درصد گیاهچههای عادی با میانگین 7/95 درصد در بذرهای تیمارشده با قارچکش دیویدند استار مشاهده گردید. بعد از گذشت 2 و14ماه از ضدعفونی بذرها وکاشت آنها در مزرعه، در مقایسه بین ارقام بیشترین درصد سبزشدن در مزرعه بعد از 2 و14ماه انبارداری متعلق به رقم یوسف بود. در مقایسه بین تیمارهای ضدعفونی بذر، بیشترین درصد ظهور گیاهچه در مزرعه بعد از 2ماه انبارداری متعلق به تیمار قارچکش دیویدند استار با میانگین 90 درصد بود، اما بعد از 14 ماه انبارداری بیشترین میزان درصد ظهور گیاهچه در بذرهای شاهد با میانگین 5/77 درصد مشاهده گردید. دو قارچکش دیویدند استار و کاربوکسین تیرام بعد از 14 ماه انبارداری بذرها، باعث کاهش در استقرار گیاهچه در مزرعه در مقایسه با بذرهای شاهدگردیدند، بهطوریکه تأثیر منفی قارچکش کاربوکسین بیشتر بود. بهطورکلی بذرها بعد از ضدعفونی (حدوداً 2 ماه) در سال اول در مزرعه کشت گردند چرا که با گذشت زمان بهمدت یکسال و بیشتر، قارچکشها و بهویژه کاربوکسین تیرام پودری دارای تأثیر منفی بر بنیه بذر و استقرار گیاهچه میباشند.
