ارزیابی صدمات فیزیکی، جوانهزنی و بنیه بذر لاینهای والدین ذرت هیبرید در ماشین دانهکن بلال
ماشین دانهکن (شیلر) بلال از تجهیزات مهم در فرایند تولید بذر ذرت بوده و عملکرد صحیح آن در کاهش میزان تلفات بذر و یکنواختی بذور تولید شده موثر میباشد. در این تحقیق صدمات فیزیکی، جوانهزنی و بنیه بذر لاینهای اینبرد والدین ذرت هیبرید دانهشده با ماشین دانهکن بلال ارزیابی شد. پارامترهای بررسیشده شامل راندمان جدا سازی بذر از بلال، راندمان تمیزش و ظرفیت عملکردی ماشین، درصد بذرهای صدمهدیده، تغییرات درصد جوانهزنی و درصد خروج ریشهچه بذرهای ذرت قبل و بعد از ماشین دانهکن بلال بودند که تغییرات میانگینهای آنها با استفاده از آزمون تی استیودنت نیز مقایسه شد. نتایج بدستآمده نشان داد که در شرایط خوراکدهی مناسب دستگاه و ظرفیت عملکردی در حدود 7/2 تن در ساعت، بذرهای بیش از 99 درصد از بلالهای ورودی دانه شد. همچنین برای بذرهای دارای رطوبت کمتر از 13 درصد، صدمات فیزیکی (شکستگی) بذر تغییرات معنیداری نداشت و کمتر از 4/0 درصد بود. میزان تمیزش بذرهای خارجشده از خروجی اصلی حدود 95 درصد و میزان بذرهای باقیمانده روی چوب بلال کمتر از 1 درصد بهدست آمد. علاوه بر موارد ذکر شده عملکرد ماشین تأثیر منفی معنیداری بر درصد جوانهزنی بذر و خروج ریشهچه نداشته و تغییرات کاهشی آنها بهترتیب کمتر از 5/0 و 2 درصد بود.

اثر زوال مصنوعی و انبارداری طبیعی بر نشت کاتیونی و قابلیت حیات بذرهای نخود ایرانی (Cicer arietinum) رقم هاشم
این تحقیق بهمنظور بررسی اثر زوال مصنوعی و انبارداری طبیعی بر خصوصیات جوانهزنی، نشت الکترولیتها و همچنین بررسی قدرت و قابلیت حیات بذرهای نخود زراعی تحت این دو شرایط و مقایسه این دو نوع زوال مصنوعی و انبارداری طبیعی در دانشگاه علوم کشاورزی و منابع طبیعی گرگان در سال 1394 اجرا شد. آزمایش بهصورت طرح کاملا تصادفی با چهار تکرار اجرا شد. تیمارها شامل 8 سطح (انبارداری طبیعی 2، 4 سال، زوال مصنوعی 1، 2، 3، 4، 5 روز و شاهد) بودند. صفات مورد بررسی در این مطالعه درصد جوانهزنی، درصد گیاهچه عادی، سرعت جوانهزنی، شاخص قدرت بذر، هدایت الکتریکی، پراکسیداسیون لیپید و میزان سدیم و پتاسیم بذرها قبل و پس از آبنوشی و همچنین مقدار آنها در محلول آبشوییشده بودند. نتایج تجزیه واریانس دادهها نشان داد اثر تیمار زوال بر همه خصوصیات جوانهزنی و نشت کاتیونی به جز میزان سدیم بذر قبل از آبشویی معنیدار بود. نتایج مقایسه میانگین دادهها نشان داد کاهش درصد و سرعت جوانهزنی، شاخص قدرت بذر و درصد گیاهچه عادی در شدتهای زوال مصنوعی 4 و 5 روز بیشتر از انبارداری طبیعی 2 و 4 سال بود. تولید مالوندیآلدئید، هدایت الکتریکی و نشت سدیم و پتاسیم در آب نیز با افزایش شدت زوال مصنوعی تا 5 روز نسبت به انبارداری طبیعی افزایش بیشتری یافت که بهدلیل عدم توانایی غشاها در حفظ انسجام خود بود. با افزایش شدت زوال میزان سدیم و پتاسیم باقیمانده در بافت بذری کاهش یافت. همچنین رنگآمیزی بذور با تترازولیوم و نیتروبلوتترازولیوم نشان داد که افزایش شدت زوال سبب کاهش قابلیت حیات بذرها شد و این کاهش قابلیت حیات در تیمارهای زوال مصنوعی شدید، بیشتر از تیمارهای انبارداری طبیعی بود. پراکسیداسیون لیپید طی زوال منجر به خسارت به غشا و افزایش نشت الکترولیتها میشود. در نتیجه قابلیت حیات بذرها کاهش یافته و منجر به کاهش درصد و سرعت جوانهزنی و کاهش درصد گیاهچه عادی تولیدی شد. این نتایج مشخص نمود که اثر انبارداری طبیعی و زوال مصنوعی بر خصوصیات جوانهزنی، نشت کاتیونی و قابلیت حیات بذرهای نخود مشابه بوده و میتوان از آزمون زوال مصنوعی در جهت برنامههای نگهداری بذرهای نخود در انبار استفاده نمود هر چند ممکن است مکانیسمهای دخیل در این دو نوع زوال با هم متفاوت باشد.

تأثیر دگرآسیبی عصاره نرگس (Narcissus tazetta L.) برخصوصیات جوانهزنی، رشدی و فیزیولوژیک اگروپیرون (Agropyron repens) و یولاف وحشی (Avena fatua)
این تحقیق با هدف بررسی اثرات دگرآسیبی عصاره آبی سوخ و برگ نرگس (Narcissus tazetta L.) بر برخی خصوصیات جوانهزنی، رشدی و فیزیولوژیکی دو علفهرز اگروپیرون (Agropyron repens) و یولاف وحشی (Avena fatua) انجام شد. برای این منظور دو آزمایش مجزا در قالب طرح کاملاً تصادفی با سه تکرار در شرایط آزمایشگاه و گلخانه در سال 1396 اجرا شد. تیمارها شامل سه سطح عصاره آبی سوخ و برگ نرگس (1، 2 و 3 درصد وزنی- حجمی) بههمراه شاهد (آب مقطر) بود. نتایج نشان داد افزایش غلظت عصاره سوخ و برگ سبب کاهش درصد و سرعت جوانهزنی بذور، طول ریشه، ارتفاع گیاه و محتوای کلروفیل a، b و کل اگروپیرون و یولاف شد ولی قندهای محلول کل و فعالیت آنتی اکسیدانت افزایش یافتند. غلظت 3 درصد عصاره سوخ مقادیر درصد جوانهزنی، طول ریشه، ارتفاع گیاه و کلروفیل کل را در گیاه اگروپیرون بهترتیب 81، 42، 38 و 49 درصد و در گیاه یولاف بهترتیب 63، 44 ، 36 و 40 درصد در مقایسه با تیمار شاهد کاهش داد. نتایج این تحقیق نشان داد که میتوان از اثرات دگرآسیبی عصاره سوخ و برگ نرگس برای کنترل بیولوژیک این دو علفهرز استفاده کرد.

اثر پلیآمینها بر جوانهزنی، بیان برخی ژنها و فعالیت آنزیمهای آنتیاکسیدانت در بذرهای کمبنیه جو
به منظور بررسی اثرات فرسودگی بذر و کاربرد پلیآمینها بر شاخصهای جوانهزنی، بیان ژن و فعالیت آنزیمهای آنتیاکسیدانت گیاهچههای جو آزمایشی بهصورت فاکتوریل در قالب طرح کاملا تصادفی در سه تکرار انجام گرفت. تیمارهای آزمایش شامل سه سطح فرسودگی (شاهد، 11 و 14 روز) و دو نوع پلیآمین (پوترسین و اسپرمین) بود. صفات اندازهگیری شامل شاخصهای جوانهزنی، فعالیت چند آنزیم آنتیاکسیدانت و بیان ژنهای مربوط به آنها بود. نتایج بررسی نشان داد که فرسودگی بذر بهمدت 14 روز موجب کاهش درصد جوانهزنی به میزان 24 درصد نسبت به شاهد شد، ولی کاربرد پوترسین آن را حدود 18 درصد افزایش داد. سرعت جوانهزنی نیز در بذرهای 14 روز فرسودهشده به میزان 36 درصد کاهش داشت که کاربرد اسپرمین آن را تا حدود 5/87 درصد افزایش داد. وزن خشک ساقهچه و ریشهچه بهترتیب در حدود 2 و 3 برابر در تیمار کاربرد پوترسین و عدم فرسودگی نسبت به شاهد بیشتر بود. بیشترین میزان فعالیت آنزیمهای آسکوربات پراکسیداز، پراکسیداز، پلیفنل اکسیداز و سوپراکسید دیسمیوتاز در بذرهای فرسوده بهمدت 14 روز با کاربرد پوترسین مشاهده شد. میزان بیان نسبی ژنهای رمزکننده آنزیمهای سوپراکسیددیسموتاز و گلوتاتیون ترانسفراز در تیمار پوترسین و بذرهای فرسوده بهمدت 14 روز، 8 و 22 برابر نسبت به شاهد بیشتر شد. همچنین، بیان نسبی ژنهای HSP90 و PR10 نیز در همین تیمار بهترتیب 5 و 8 برابر شاهد بود. بهطورکلی، کاربرد پوترسین تا حدود زیادی موجب بهبود قدرت بذر جو گردید که این امر میتواند ناشی از بهبود شرایط اکسیداتیو سلول، مقاومت پروتئینهای بذر به دلیل فعالیت بیشتر HSP90 و افزایش غلظت درونزاد هورمونهای بذر باشد.

بررسی برخی خصوصیات مرتبط با قابلیت جوانهزنی و بنیه گیاهچه بادامزمینی رقم NC2 در بذرهای حاصل از بوتههای محلولپاشیشده با متانول و متیلوباکتریوم
بهمنظور بررسی خصوصیات مرتبط با قابلیت جوانهزنی و بنیه گیاهچه بادامزمینی در بذرهای حاصل از بوتههای محلولپاشیشده با متانول و متیلوباکتریوم تحقیقی آزمایشگاهی در 2 سال در دانشگاه آزاد اسلامی واحد رشت به اجرا درآمد. این تحقیق با استفاده از آزمونهای جوانهزنی استاندارد، سرما و پیری تسریعشده اجرا شد. هر یک از آزمونها با استفاده از آزمایش فاکتوریل بر پایه طرح بلوکهای کامل تصادفی با 16 تیمار (جمعیت باکتری متیلوباکتریوم بر حسب CFU در 4 سطح صفر، 106، 108 و 1012 و 4 سطح مقدار متانول صفر، 10، 20 و 30 درصد حجمی) در 3 تکرار اجرا شد. خصوصیات مورد بررسی عبارت بودند از درصد جوانهزنی نهایی، بنیه گیاهچه، وزن خشک ریشهچه، وزن خشک هیپوکوتیل، وزن خشک ساقهچه و وزن خشک گیاهچه. نتایج نشان داد که اثر سال بر تمام خصوصیات مورد بررسی در کلیه آزمونها معنیدار بود. جمعیت باکتری فقط اثر معنیداری بر درصد جوانهزنی نهایی در کلیه آزمونها داشت. اما مقدار متانول اثر معنیداری بر تمام خصوصیات مورد بررسی در هر سه آزمون داشت. اثر متقابل جمعیت باکتری در مقدار متانول نیز فقط بر درصد جوانهزنی نهایی در آزمونهای جوانهزنی استاندارد و سرما اثر معنیداری داشت. مقایسه میانگین صفات مورد بررسی نیز نشان داد که بیشترین مقدار درصد جوانهزنی نهایی در کلیه آزمونها در جمعیت باکتری CFU 1012 مشاهده شد. بیشترین مقدار تمام خصوصیات بررسیشده در کلیه آزمونها از مقدار 20 درصد حجمی متانول بهدست آمد.

مقایسه آزمونهای هدایت الکتریکی، آبنوشی و هیلتنر برای پیش بینی سبزشدن گیاهچه عدس در مزرعه
آزمونهای مختلفی برای تعیین قدرت بذر معرفی شدهاند که در هر گونه گیاهی، یک یا برخی از آنها مناسب هستند. این پژوهش برای مقایسه آزمونهای هدایت الکتریکی، آبنوشی و هیلتنر در تعیین قدرت بذر عدس اجرا گردید. این تحقیق در دو بخش آزمایشگاه و مزرعه در دانشگاه رازی و در سال زراعی 94-1393 انجام شد. در مزرعه، هشت توده بذر عدس که از نظر رقم و سن متفاوت بودند کشت شدند. در آزمایشگاه نیز این هشت نمونه بذر در آزمونهای هدایت الکتریکی، آبنوشی و هیلتنر مورد مقایسه قرار گرفتند. نتایج آزمایش مزرعهای نشان داد که نمونههای بذر از نظر صفتهای درصد و سرعت سبزشدن، میانگین سبزشدن روزانه و متوسط زمان لازم برای سبزشدن دارای اختلاف معنیدار بودند. تحقیق آزمایشگاهی مشخص نمود که در آزمونهای هدایت و هیلتنر، بین نمونههای بذر اختلاف معنیدار وجود داشت اما از نظر آزمون آبنوشی، اختلافی بین نمونهها مشاهده نشد. در تجزیه گروهی در آزمونهای هدایت و هیلتنر، نمونههای بذر جدیدتر از قدرت بالاتری برخوردار بودند. بر اساس تجزیه همبستگی، آزمونهای هدایت و آبنوشی با درصد و سرعت سبزشدن گیاهچه در مزرعه همبستگی نداشتند اما صفت تعداد گیاهچه طبیعی در آزمون هیلتنر دارای همبستگی مثبت معنیدار با درصد سبزشدن (r=0.63) و سرعت سبزشدن (r=0.498) گیاهچه در مزرعه بود. بنابراین، بهنظر میرسد که برای پیشبینی سبزشدن گیاهچه در مزرعه، آزمونهای هدایت و آبنوشی دارای قابلیت نامناسب و آزمون هیلتنر دارای قابلیت مناسب باشند.

اثر محلولپاشی، نوع کود و زمان مصرف بر رشد و تولید بنه توده بومی زعفران (Crocus sativus L.) تربت حیدریه
عامل اصلی محدودکننده کشت زعفران (Crocus sativus L.) سختی و دشواری بهدست آوردن بنههایی با کیفیت بالا برای تکثیر است. لذا این تحقیق با هدف مقایسه اثر محلولپاشی سه نوع کود در زمانهای مختلف بر رشد و تولید بنه در زعفران بهصورت کرتهای خردشده در قالب بلوکهای کامل تصادفی با سه تکرار در سال زراعی 93-1392 در مزرعهای در شهرستان تربت حیدریه خراسان رضوی اجرا شد. تیمار اصلی شامل کودهای دلفان پلاس، اولتراسول 1، اولتراسول 2 و شاهد (آبپاشی برگها بدون عناصر غذایی و معادل حجم کود در زمان کودپاشی)، تیمار فرعی سه زمان مختلف کاربرد شامل 10 اسفند، 5 فروردین و 25 فروردین بود. صفات اندازهگیری شامل وزن خشک و قطر بنههای تولیدی و تعداد کل بنههای مادری و تولیدی بود. نتایج اثرات تیمارها و زمانهای مختلف محلولپاشی بر اندازه، وزن بنههای تولیدی معنیدار بود، اما بر تعداد کل بنههای تولیدی در واحد سطح و همچنین برهمکنش تیمارها معنیدار نبود. در مجموع، نتایج این پژوهش، برتری همه کودها نسبت به تیمار شاهد و کود دلفان پلاس نسبت به سایر کودها و همچنین نسبت به تیمار شاهد (بهترتیب 5/13، 6/13 و13 درصد) را در افزایش اندازه، وزن بنهها نشان داد. در بین تیمارهای زمان محلولپاشی، تاریخ 5 فروردین نسبت به سایر تاریخها بر اندازه، وزن بنه برتری داشت. بهطوریکه زمان محلولپاشی 5 فروردین نسبت به دو زمان 10 اسفند و 25 فروردین باعث افزایش 6/6 و 11 درصدی در اندازه بنههای تولیدی شد. بنابراین در بین کودهای استفاده شده، کود دلفان پلاس بهترین کود بود و در بین زمانهای مصرف نیز، 5 فروردین مناسبترین زمان مصرف کود بود.

بررسی ارقام جو در شرایط تنش شوری در مراحل رشد اولیه گیاهچه با استفاده از تجزیه چندمتغیره
افزایش تحمل به شوری در جو نیاز اساسی جهت پایداری تولید این محصول در مناطق شور محسوب میشود. تحقیق حاضر بهمنظور مطالعه تنوع ژنتیکی در 20 رقم جو زراعی در شرایط تنش شوری جهت شناسایی متحملترین ارقام در مراحل جوانهزنی و رشد اولیه گیاهچه براساس صفات مورفولوژیک و فیزیولوژیک انجام شد. این تحقیق در سال 1394 در آزمایشگاه تحقیقاتی دانشگاه آزاد اسلامی واحد اهواز در مراحل رشد اولیه گیاهچه بهصورت آزمایش فاکتوریل در قالب طرح کاملاً تصادفی با 3 تکرار شامل 20 رقم جو زراعی بهعنوان عامل اول و تنش شوری با کلرید سدیم در 4 سطح صفر (آب مقطر)، 5، 10 و 15 دسیزیمنس بر متر بهعنوان عامل دوم اجرا گردید. نتایج تجزیه واریانس دادهها نشان داد که بین ارقام از نظر اغلب صفات مورد مطالعه تفاوت معنیداری وجود داشت که نشاندهنده وجود تنوع کافی بین ارقام میباشد. تجزیه به مؤلفههای اصلی، 11 صفت بررسیشده را به 2 مؤلفه تقسیم و بر اساس آن 62 درصد از کل تغییرات دادهها را توجیه نمود. تجزیه خوشهایی ارقام را به 3 خوشه تقسیم نمود. از جمعبندی نتایج تجزیه به مولفههای اصلی، نمودار بای پلات و تجزیه خوشهای مشخص شد که ارقام گرگان، کویر، بهرخ، ترکمن و زرجو تحمل بهتری به تنش شوری نسبت به سایر ارقام داشتند و در مرحله بعدی ارقام فجر30، صحرا، ریحان 48 و کارون قرار گرفتند.

این تحقیق بهمنظور بررسی تأثیر محتوای رطوبت بذر و مدت انبارکردن بر برخی خصوصیات جوانهزنی گیاهچه سه رقم گندم در استان مازندران بهصورت کرتهای دو بار خرد شده بر اساس طرح بلوکهای کامل تصادفی با 4 تکرار اجرا شد. تیمارها بذرهای ارقام مروارید، میلان و N8019 برداشتشده با محتوای رطوبتهای10،12 ،14 و16درصد و انبارشده بهمدت 2،3 و4 ماه بعد از برداشت بودند. با آزمون جوانهزنی استاندارد، درصد جوانهزنی نهایی، درصد گیاهچههای عادی، متوسط زمان جوانهزنی، طول و وزن خشک گیاهچه و شاخصهای وزنی و طولی بنیه (بهترتیب حاصلضرب وزن خشک و طول گیاهچه در درصد جوانهزنی نهائی) گیاهچه تعیین و هدایتالکتریکی مواد مترشحه از بذرها اندازهگیری شدند. نتایج نشان داد بذرهای برداشتشده با محتوای رطوبت10 درصد و انبارشده بهمدت2 ماه، بذرهای ارقام میلان و N8019انبارشده بهمدت4 ماه و بذرهای رقم میلان برداشتشده با محتوای رطوبت12 درصد از بیشترین درصد جوانهزنی نهائی بهمیزان 98 درصد برخوردار بودند. بذرهای ارقام مروارید و میلان برداشتشده با محتوای رطوبت10 درصد و 2 ماه انبارشده و بذرهای رقم N8019با محتوای رطوبت 12 درصد برداشت و 2 ماه انبارشده دارای بیشترین درصد گیاهچههای عادی بهمیزان 98 درصد بودند. کمترین متوسط زمان جوانهزنی به بذرهای رقم میلان برداشتشده با محتوای رطوبت بذر10 درصد و 2 ماه انبارشده بهمیزان210/1 روز، تعلق داشت. بر اساس نتایج این تحقیق، محتوای رطوبت بذر در هنگام برداشت مطلوب برای رقم مروارید10 و برای ارقام میلان و N8019 12 درصد و مدت انبارمانی مناسب بذر برای رقم مروارید سه ماه و برای ارقام میلان و N8019 دو ماه بود.

مقاله مروری: راهبردهای تلفیقی سلامت بذر برای پیشگیری از دژنراسیون بذر
مواد تکثیری سالم و اصیل از مهمترین نهادهها برای تولید سالم و مقرون بهصرفه محصولات کشاورزی هستند. با توجه به نوع میزبان، گونه / نژاد بیمارگر یا آفت، شرایط محیطی و ... بذر ناقل بسیاری از بیمارگرها و آفات کشاورزی به نسل بعد محسوب میشود. اهمیت این موضوع بهویژه در مورد مواد تکثیری غیرجنسی بهدلیل انتقال کامل بیمارگرهای موجود در گیاهان مادری به نتاج و تجمع آنها در طی کشتهای متوالی در صورت استفاده از بذور غیر رسمی/گواهینشده نمود بیشتری دارد. در گیاهانی که بهصورت غیرجنسی تکثیر مییابند، استفاده مداوم از مواد تکثیری سالهای گذشته برای کشت، تجمع آفات و بیمارگرها را در پی دارد که نتیجه آن کاهش مستمر کمیت، کیفیت و بازارپسندی محصول است. این پدیده که دژنراسیون بذر (ماده تکثیری) نام دارد، در گیاهانی که از طریق غده، پیاز و ریزوم تکثیر مییابند، نمود بارزتری دارد. دژنراسیون بذر عموما در مناطقی اتفاق میافتد که تولیدکنندگان به هر دلیل به مواد تکثیری سالم و گواهیشده دسترسی نداشته و مجبور به استفاده از محصول زراعی سال قبل یا بذور غیر رسمی بهعنوان ماده تکثیری هستند. بهدلیل تاثیر فاکتورهای مختلف زراعی، بیولوژیک و محیطی در بروز دژنراسیون مواد تکثیری، کنترل تلفیقی که تاثیر این قبیل فاکتورها را به حداقل برساند، برای پیشگیری یا کاهش اثر این پدیده نیاز است. در مقاله حاضر به جزئیات پدیده دژنراسیون در مواد تکثیری، فاکتورهای مختلف تاثیرگذار بر بروز این پدیده و نیز روشهای تلفیقی در پیشگیری از آن پرداخته میشود.
