بررسی تاثیر پرایمینگ بذر بر جوانه‌زنی و صفات مورفوفیزیولوژیک نشاءهای برنج رقم هاشمی(Oryza sativa L.cv. Hashemi) تحت رژیم‌های‌مختلف‌رطوبتی

چکیده

دو آزمایش مستقل به­صورت فاکتوریل در قالب طرح کاملاً تصادفی در پژوهشکده علوم گیاهی دانشگاه فردوسی مشهد انجام شد. در آزمایش اول سطوح رژیم رطوبتی شامل صفر، 2-، 4-، 6-، 8- و 10- بار بر جوانه­زنی بذور برنج با سه سطح پیش­تیمار شامل بدون پیش­تیمار (شاهد)، هالوپرایمینگ با کلریدکلسیم و هیدروپرایمینگ مورد بررسی قرار گرفت. در آزمایش دوم بذرهای پیش­تیمارشده مشابه آزمایش اول, در گلدان­های حاوی ماسه کشت و پنج رژیم رطوبتی شامل شاهد (ظرفیت زراعی), 20، 40، 60 و 80 درصد ظرفیت زراعی بر روی آن­ها اعمال شد. پس از 28 روز، برخی از صفات مورفوفیزیولوژیک اندازه­گیری شد. نتایج آزمایش اول نشان داد که رژیم­های مختلف رطوبتی، صفات جوانه­زنی را به­طور منفی تحت تاثیر قرار داد. پرایمینگ بذر سبب بهبود سرعت و درصد جوانه­زنی بذر، وزن خشک ساقه­چه و ریشه­چه در برخی از رژیم­های رطوبتی شد. کاربرد هیدروپرایمینگ نتایج بهتری را نسبت به هالوپرایمینگ نشان داد. در آزمایش دوم سطوح پایین رطوبتی به­صورت معنی­داری ارتفاع نشاء، وزن خشک بخش هوایی، سطح برگ، سطح و قطر ریشه، محتوای نسبی آب و شاخص پایداری غشا را کاهش داد. در این شرایط استفاده از پرایمینگ تاثیر مثبتی بر صفات فوق نداشت، ولی مقاومت روزنه­ای و میزان کلروفیل برگ را افزایش داد. نتایج این ازمایش نشان داد که هم در شرایط بدون تنش و هم در سطوح پایین رطوبتی،کاربرد پرایمینگ (به­خصوص هیدروپرایمینگ) تاثیر مثبت و معنی­داری بر صفات مرتبط با جوانه­زنی بذر برنج دارد.

چکیده [English]

Two separate factorial experiments based on the completely randomized design were conducted at the Research Center for Plant Sciences, Ferdowsi university of Mashhad. In the first experiment, the effect of 0, -2, -4, -6, -8, and -10 bar of moisture regimes with three priming levels including control (without priming), halo priming by calcium chloride, and hydropriming were investigatedon the seed germination of rice. In the second experiment, the seeds primed similar to the first experiment were cultivated in the pots filled with sand, being under five levels of moisture regimes including control (filed capacity), 20, 40, 60, and 80% of field capacity. After 28 days of cultivation, some morphophysiological traits were measured. The results of the first experiment showed that under low moisture regimes the germination traites were negatively affected. However, under some moisture regimes, the use of priming improved the germination rate and percentage, and increased dry weight of radicle and hypocotyl. It was also found that improving effects of hydropriming was more than halopriming. In the second experiment, low moisture regimes significantly decreased seedling height, shoot dry weight, leaf area, root area and diameter, water relative content, and membrane stability index. Under this condition, seed priming treatment did not positively affect these traits, butincresedstomatal resistance and chlorophyl conten.According to the results, it seems that both under control and low moisture regimes the use of seed priming, specially hydropriming, can be beneficial for Riceseed germination.
