تأثیر اسید آسکوربیک بر جوانه‌زنی و فعالیت آنزیم‌های آنتی اکسیدان در بذرهای زوال یافته گلرنگ رقم گلدشت

چکیده

به منظور بررسی تأثیر اسید آسکوربیک بر جوانه‌زنی و ویژگی­های فیزیولوژیک بذرهای زوال یافته گلرنگرقم گلدشت، آزمایشی به صورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار اجرا گردید. بذرهای بدون زوال و زوال یافته گلرنگ (نگهداری در دمای 40 درجه سانتی­گراد به مدت 4،6، 8 و 10 روز) با غلظت­های 0، 28/0 و 56/0میلی مولار اسید آسکوربیک پیش تیمار شدند. در این پژوهش صفات درصد و سرعت جوانه­زنی، بنیه گیاهچه، طول ساقه‌چه و ریشه‌چه، وزن خشک ساقه‌چه و ریشه‌چه، میزان مالون دی آلدئید، فعالیت آنزیم‌های کاتالاز، سوپراکسید دیسموتاز و میزان پروتئین اندازه­گیری شد. نتایج نشان داد با افزایش زوال جوانه­زنی و رشد گیاهچه کاهش یافت. در تمامی سطوح زوال‌‌، غلظت 28/0 میلی مولار اسید آسکوربیک سرعت جوانه‌زنی، بنیه گیاهچه، طول ساقه‌چه و ریشه‌چه و وزن خشک ساقه‌چه و ریشه‌چه را نسبت به سایر غلظت‌ها افزایش داد. زوال میزان مالون دی آلدئید را افزایش داد ولی فعالیت آنزیم‌های کاتالاز، سوپراکسید دیسموتاز و میزان پروتئین را کاهش داد. در تمامی سطوح زوال، اسید آسکوربیک 28/0 میلی مولار سبب کاهش میزان مالون دی‌آلدئید بعد از 12 ساعت آبگیری شد، در حالی­که فعالیت کاتالاز، سوپراکسید دیسموتاز و میزان پروتئین را افزایش داد. نتایج این آزمایش نشان داد غلظت  28/0میلی مولار اسید آسکوربیک در بذرهای زوال یافته گلرنگ توانست جوانه­زنی و رشد گیاهچه را با کاهش پراکسیداسیون غشاء و افزایش فعالیت آنزیم کاتالاز، سوپراکسید دیسموتاز بعد از 12 ساعت آبگیری بهبود بخشد

In order to study the effect of ascorbic acid on germination and some physiologic characteristics of aged safflower an experiment was conducted as factorial arrangement in completely randomized design with three replications. Non-aged and aged (40 °C for 4, 6, 8 and 10 d) seeds of safflower were subjected to priming with ascorbic acid at concentrations of 0, 0.28 and 0.56 mM. In this research germination percentage and rate, seedling vigour, shoot and root length, shoot and root dry weight, malondialdehyde, catalase activity, superoxide dismutase activity and protein content measured. The results showed that germination and seedling growth decreased with aging increment. In all levels of aging, germination percentage and rate, shoot and root length, shoot and root dry weight increased at 0.28 mM ascorbic acid than other concentrations. Accelerated aging not only resulted in increased malondialdehyde, but also in decreased catalase activity, superoxide dismutase activity and protein content. Priming with 0.28 mM ascorbic acid decreased malondialdehyde after 12 hours imbibition, whereas increased catalase activity, superoxide dismutase activity and protein content under aging. The results suggest that aged safflower seeds priming with 0.28 mM ascorbic acid improves the germination and seedling growth by reducing membrane peroxidation and increasing the activities of catalase and superoxide dismutase enzymes after 12 hours imbibition
