ارزیابی تاثیر تنش شوری بر فعالیت برخی آنزیم های آنتی اکسیدان در گیاهچه های حاصل از بذر پرایم شده ماریتیغال (Silybum marianum)

چکیده

این پژوهش به­منظور بررسی فعالیت آنزیم­های آنتی­اکسیدان در گیاهچه­های حاصل از بذر پرایم­شده ماریتیغال در شرایط تنش شوری انجام گرفت. آزمایش به­صورت فاکتوریل با چهار فاکتور و سه تکرار در قالب طرح کاملا تصادفی در پژوهشکده بیوتکنولوژی کرج اجرا شد. فاکتور اول: ژنوتیپ ( رقم مجارستان و توده بومی آمل)، فاکتور دوم: بذر پرایم­شده و بدون پرایم، فاکتور سوم: تنش شوری (150 میلی­مولار) و شاهد (صفر میلی­مولار)، و فاکتور چهارم: مراحل نمونه­برداری ( 24 و 96 ساعت پس از کاشت) بود. نتایج نشان داد که پرایمینگ بذر باعث افزایش معنی­دار فعالیت آنزیم کاتالاز در رقم مجارستان شد، به­طوری­که فعالیت کاتالاز در بذور پرایم­شده نسبت به شاهد به میزان 96 درصد افزایش یافت. همچنین پرایمینگ فعالیت آنزیم پراکسیداز را در هر دو ژنوتیپ و آسکوربات پراکسیداز را در توده بومی آمل افزایش داد. افزایش فعالیت آنزیم­های کاتالاز و آسکوربات پراکسیداز در بذور پرایم­شده در مرحله جوانه­زنی و افزایش فعالیت پراکسیداز در مرحله آبنوشی مشاهده گردید. به­طور­کلی نتایج نشان داد در شرایط تنش شوری فعالیت آنزیم­های آنتی­اکسیدان کاتالاز و پراکسیداز و آسکوربات پراکسیداز برای بذور پرایم­شده بیشتر از بذور بدون پرایم بود، همچنین پرایمینگ در رقم مجارستان فعالیت آنزیم­های کاتالاز و پراکسیداز را بیش­تر از توده آمل تحت تاثیر قرار داد.

چکیده [English]

This experiment was performed in order to evaluation antioxidant enzymes activity in seedling induced of milkthistle primed seeds in salt stress condition. The experiment was conducted in factorial based on completely randomized design with four factors and three replications in Agricultural Biotechnology Research Institute of Iran (ABRII). These factors were included of genotype (Amol and Majar), priming (primed and non-primed seeds), stress: salinity (150 mM) and control led (non-salinity) and time of sampling in 24 and 96 hours after sowing. The results showed the seed priming increased significantly the catalase (CAT) activity in Majar , proxidase(POX) activity in both genotypes and ascorbat peroxidase (APX) in Amol, so that CAT activity increased in Majar primed seeds than control 96%. Priming increased POX activity in Majar 14-fold greater than Amol. Increasing CAT and APX activity were observed in primed seeds in germination stage but POX activity increasing was observed in imbibition stage. Overall, the results showed the CAT, POX and APX activity was higher in primed seeds than non-primed in salt stress condition. Priming affected CAT and POX activity in Majar greater in Amol.
