تعیین بهترین روش و مدت زمان پیش تیمار بذر در گیاه دارویی سرخارگل (Echinacea purpurea) در شرایط آزمایشگاهی و گلدانی
بهمنظور تعیین بهترین روش و مدتزمان پیشتیمار بذر بر ویژگیهای گیاهچهای گیاه سرخارگل در شرایط آزمایشگاهی و گلدانی، دو آزمایش بهصورت مجزا در دانشکده کشاورزی دانشگاه ارومیه به اجرا درآمد. هر دو آزمایش در قالب فاکتوریل بر پایه طرح کاملاً تصادفی اجرا شدند که روشهای مختلف پیشتیمار بذر (هاردنینگ، هیدروترمال، هیدروپرایم، کلریدپتاسیم، نیتراتکلسیم، کلریدسدیم، نیتراتپتاسیم و شاهد) و مدتزمان (3 و 6 ساعت) تیمارهای آزمایشی بودند. نتایج حاصل از تجزیهواریانس نشان داد که اختلاف کاملاً معنیداری بین روشهای مختلف پرایمینگ چه در شرایط آزمایشگاهی و چه در شرایط گلدانی وجود داشت ولی بین مدتزمان پرایمینگ در شرایط آزمایشگاهی هیچگونه اختلافی مشاهده نشد. با این حال با افزایش مدتزمان در شرایط گلدانی، تأثیر مثبت پیشتیمار بر مؤلفههای جوانهزنی بیشتر شد. نکته قابلتوجه در این تحقیق این بود که تمامی روشهای پیشتیمار باعث بهبود مؤلفههای جوانهزنی نمیشوند، بهطوریکه کلریدسدیم در شرایط آزمایشگاهی و هیدروترمال در شرایط گلدانی باعث کاهش درصد و سرعتجوانهزنی در مقایسه با شاهد شدند. بهطور کلی روشهای مختلف پیشتیمار، تأثیر مثبتی بر ویژگیهای جوانهزنی گیاه سرخارگل و پیشتیمارهای هاردنینگ و کلریدپتاسیم در مدتزمان 6 ساعت بیشترین تأثیر را بر صفات درصد، سرعت و قدرتجوانهزنی داشتند. بنابراین میتوان از این پیشتیمارها جهت بهبود قدرت و کیفیتبذر گیاه سرخارگل بهمنظور افزایش تحمل گیاه به شرایط نامساعد محیطی و افزایش یکنواختی و سطح سبزمزرعه استفاده نمود.

شناسایی و بررسی پتانسیل آللوپاتیک آلکالوئیدهای تاتوره (Datura stramonium) بر ویژگیهای جوانهزنی ارقام ذرت
به منظور شناسایی آلکالوئیدهای موجود در علف هرز تاتوره، عصاره شاخساره آن تهیه و با استفاده از روش گاز کروماتوگرافی تجزیه شد. سه آلکالوئید تروپینون، تروپین و آتروپین که دارای پتانسیل آللوپاتیکی می باشند، شناسایی و استخراج گردید. پتانسیل آللوپاتیک آلکالوئیدهای مذکور بر جوانه زنی و رشد گیاهچه شش رقم ذرت شامل سینگل کراسهای301، 404، 500، 647، 700 و 704 به صورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار مورد بررسی قرار گرفت. نتایج نشان داد که اثر متقابل آلکالوئید×غلظت×رقم بر درصد و سرعت جوانه زنی معنی دار بود. بیشترین پتانیسل بازدارندگی مربوط به تروپینون و کمترین آن مربوط به تروپین بود. با افزایش غلظت آلکالوئیدها اثر بازدارندگی نیز افزایش یافت. ویژگی های وزن خشک ریشه چه و ساقه چه تمامی ارقام ذرت نیز تحت تأثیر پتانسیل بازدارندگی تروپینون و آتروپین قرار گرفتند. هر دو صفت مذکور در همه ارقام در معرض تروپین افزایش یافتند. نتایج کلی بیانگر این است که در عصاره ی بقایای علف هرز تاتوره سه آلکالوئید با اثرهای متفاوت تحریککننده و بازدارنده وجود دارد، و همچنین غلظت های مختلف هر آلکالوئید نیز از پتانسیل آللوپاتیکی متفاوتی برخوردار است که می توانند رشد گیاه زراعی را تحت تأثیر خود قرار دهند. یافتن مکانیسم های اثر بازدارندگی این ترکیبها بر گیاهان دیگر ممکن است برای یافتن ترکیباتی که بتوانند به عنوان علفکش مورد استفاده قرار گیرند، مفید واقع شوند.

Merr.] با ظهور گیاهچه در مزرعه، پژوهشی آزمایشگاهی و مزرعه ای در مؤسسه تحقیقات ثبت و گواهی بذر و نهال در کرج به اجرا درآمد. آزمایش به صورت فاکتوریل بر پایه طرح بلوک های کامل تصادفی با 12 تیمار [سه قابلیت جوانه زنی اولیه (قوه نامیه)، استاندارد (85 درصد)، بالای استاندارد (بالاترین میزان قوه نامیه نمونه های هر رقم) و پائین استاندارد (پایین ترین میزان قوه نامیه نمونه های هر رقم)] و 4 رقم تجاری سویا [ویلیامز، ساری (JK) تولید استان مازندران، تلار و 033 تولید استان گلستان] و چهار تکرار اجرا شد. طول گیاهچه، وزن خشک گیاهچه، درصد گیاهچه عادی، درصد گیاهچه غیرعادی، درصد جوانه زنی نهایی، متوسط زمان جوانه زنی، ضریب سرعت جوانه زنی، متوسط جوانه زنی روزانه، سرعت جوانه زنی روزانه، شاخص طولی بنیه و وزنی بنیه بذرها با آزمون جوانه زنی استاندارد و تجزیه و تحلیل رشد گیاهچه اندازهگیری شدند. همچنین بذرها در مزرعه کشت شدند و درصد ظهور اولیه و نهایی گیاهچه، سرعت ظهور و سرعت ظهور تجمعی گیاهچه، شاخص بنیه و وزن خشک گیاهچه در مزرعه تعیین شدند. نتایج نشان داد طول و وزن خشک گیاهچه، ضریب سرعت جوانه زنی، شاخص های طولی و وزنی بنیه بذر، درصد ظهور اولیه و نهایی گیاهچه و شاخص بنیه گیاهچه در مزرعه تحت تأثیر برهم کنش رقم × قابلیت جوانه زنی اولیه قرار گرفت و سایر شاخص های بررسی شده تحت تأثیر هریک از عوامل قرار گرفتند. همچنین افزایش وزن طول و خشک گیاهچه، درصد گیاهچه های عادی، سرعت جوانه زنی روزانه و شاخص طولی بنیه بذر در بذرهای با قابلیت جوانه زنی اولیه بالاتر مشاهده شد و این بذرها از درصد ظهور اولیه و نهایی و سرعت ظهور گیاهچه در مزرعه بالاتری برخوردار بودند و گیاهچه های با بنیه قوی تری داشتند. بهطورکلی بذ های رقم های ویلیامز و 033 با قابلیت جوانه زنی اولیه بالای استاندارد از لحاظ بیشتر شاخص های مورد مطالعه در آزمایشگاه و مزرعه در مقایسه با سایر رقم ها برتری معنی داری داشتند. درصد گیاهچه های عادی نسبت به درصد جوانه زنی نهایی در آزمون جوانه زنی استاندارد با ظهور نهایی مزرعه ای از همبستگی بالاتری برخوردار بود. بنابراین به نظر می رسد تعیین درصد گیاهچه عادی می تواند معیار مناسبی از بنیه بالقوه بذرها برای استقرار گیاهچه ها در مزرعه باشد.

اثر تلفیق تیمار زیستی بذر و کود شیمیایی بر عملکرد و جذب عناصر غذایی شلغم علوفهای (Brassica rapa L.)
به منظور بررسی تلفیق تیمار زیستی بذر و کود شیمیایی بر عملکرد و جذب عناصر غذایی شلغم علوفه ای، آزمایشی در مزرعه پژوهشی موسسه تحقیقات برنج کشور (رشت) در قالب طرح بلوک های کامل تصادفی در دو سال زراعی 1389 و 1390 انجام شد. تیمارهای مورد بررسی در این تحقیق شامل(1) شاهد (بدون کود و بدون تلقیح)، (2) کود کامل شیمیایی و بدون تلقیح، (3) تلقیح با ازتوباکتر کروکوکوم سویه 12+ بدون کود شیمیایی نیتروژن، (4) تلقیح با ازتوباکتر کروکوکوم سویه 12+ 100 درصد کود شیمیایی نیتروژن، (5) تلقیح با سودوموناس فلورسنس سویه 41+ بدون کود شیمیایی فسفر، (6) تلقیح با سودوموناس فلورسنس سویه 41 + 100 درصد کود شیمیایی فسفر، (7) تلقیح با ازتوباکتر کروکوکوم سویه 12+ سودوموناس فلورسنس سویه 41+ بدون کود شیمیایی نیتروژن و فسفر، (8) تلقیح بذر با ازتوباکتر کروکوکوم سویه 12+ سودوموناس فلورسنس سویه 41+ 50 درصد کود شیمیایی نیتروژن و فسفر، (9) تلقیح با ازتوباکتر کروکوکوم سویه 12+ سودوموناس فلورسنس سویه 41+ 100 درصد کود شیمیایی نیتروژن و فسفر بودند. ویژگی های مورد بررسی شامل ارتفاع بوته، قطر ساقه، میزان عناصر (فسفر و نیتروژن) در بافت گیاه، وزن خشک برگ و غده و عملکرد علوفه تر و خشک بودند. نتایج نشان داد که تلفیق تیمار زیستی بذر و کود شیمیایی اثر معنی داری بر ویژگی های مورد مطالعه داشت. همچنین اثر این تلفیق بر درصد نیتروژن، فسفر و عملکرد علوفه خشک معنی دار بود. تلقیح با ازتوباکتر کروکوکوم سویه 12+ سودوموناس فلورسنس سویه 41 به همراه سطح کودی 50 درصد و100 درصد باعث افزایش وزن خشک اندام هوایی و ریشه شد. درصد فسفر و نیتروژن بافت گیاهی در تیمارهای تلفیقی نسبت به شاهد افزایش یافت. نتایج آزمایش نشان داد که مدیریت تلفیقی کود نقش بهسزایی در افزایش عملکرد کمی و جذب عناصر غذایی شلغم علوفه ای دارد و تلقیح بذر با ازتوباکتر کروکوکوم سویه 12+ سودوموناس فلورسنس سویه 41 و 50 درصد کود شیمیایی مناسب ترین روش کوددهی شلغم علوفه ای می باشد.

تأثیر جنس کیسه، شرایط و طول دوره انبارداری بر ویژگیهای جوانهزنی بذر در آزمایشگاه و ظهور مزرعهای گیاهچه سویا
به منظور بررسی اثر شرایط و طول دوره انبارداری و همچنین جنس کیسه بر کیفیت بذر سویا(Glycine max L.) رقم ساری (JK) پژوهشی در سال های 89-1388 و 90-1389 در دو مرحله آزمایشگاهی و مزرعه ای براساس آزمایش عاملی (فاکتوریل) 2×3×5 با سی تیمار بر پایه طرح کاملاً تصادفی (بخش آزمایشگاهی) و طرح پایه بلوک های کامل تصادفی (بخش مزرعه ای) با سه تکرار در مرکز تحقیقات کشاورزی ساری انجام شد. عوامل مورد بررسی، پنج نوع لفاف، پاکت کاغذی دو لایه، پاکت کاغذی سه لایه، پاکت کاغذی دو لایه با لایه لمینت، کیسه پارچه ای (چتایی)، کیسه پروپیلنی، سه مدت نگهداری 2، 4 و 6 ماه و دو شرایط نگهداری، انبار معمولی و نگهداری در انبار کنترلشده با دمای 16 درجه سانتی گراد و رطوبت نسبی 65 درصد بود. نتایج نشان داد که با افزایش طول دوره انبارداری از دو ماه به شش ماه، میزان جوانه زنی از 87/84 درصد به 25/79 درصد کاهش یافت و در سال اول میزان جوانه زنی در انبار کنترل شده با پاکت کاغذی سه لایه (62/88 درصد) نسبت به انبار معمولی با کیسه پروپیلنی (94/63 درصد) بیشتر بود. آزمون پیری تسریع شده نشان داد که با افزایش طول دوره انبارداری از دو ماه به شش ماه میزان جوانه زنی از 81/81 درصد به 61/76 درصد کاهش یافت. در هر دو سال بیشترین درصد جوانه زنی مربوط به کیسه های کاغذی سه لایه بود و کمترین آن در کیسه های پروپیلنی و پارچه ای بود. شاخص های کیفی بذر در هر دو سال در انبار کنترل شده نسبت به انبار معمولی از شرایط بهتری برخوردار بود. نتایج مزرعه ای نیز نشان داد که بیشترین درصد ظهور نهایی گیاهچه ها در هر دو سال از بذرهای انبار کنترل شده در پاکت کاغذی سهلایه (98/81 در سال اول و 89/80 درصد در سال دوم) بود و کمترین میزان آن در هر دو سال در انبار معمولی در کیسه پروپیلنی (31/62 درصد در سال دوم و 08/62 درصد در سال اول) بود. به طور کلی مشخص شد که نگهداری بذرها در پاکت های کاغذی سهلایه در انبار کنترل شده برای نگهداری بذرهای سویا مناسب می باشد.

مدلسازی هیدروتایم جوانهزنی بذر علفهای هرز فالاریس (Phalaris minor)، تاجخروس ریشهقرمز (Amaranthus retroflexus) و تاجخروس خوابیده (A. blitoides)
مدل سازی جوانه زنی با استفاده از مدل های هیدروتایم و هیدروترمال تایم به طور گسترده ای انجام می شود. تنوع در زمان جوانه زنی ناشی از تنوع پتانسیل آب پایه بذرهای یک جمعیت است که به طور معمول توسط توزیع نرمال مدل سازی می شود. در این آزمایش، فرض نرمال بودن توزیع پتانسیل آب پایه با جوانه زنی بذر علف های هرز فالاریس (Phalaris minor)، تاج خروس ریشه قرمز (Amaranthus retroflexus) و تاج خروس خوابیده (A. blitoides) در محدوده ای از پتانسیل های آب (صفر، 2/0-، 4/0-، 6/0- و 8/0- مگاپاسکال) مورد آزمون قرار گرفت. سه توزیع آماری نرمال، ویبول و گامبل برای نشان دادن تنوع نسبی پتانسیل آب پایه جمعیت های بذری مورد مقایسه قرار گرفت. نتایج نشان داد که پارامت های برآورد شده با مدل هیدروتایم توسعه یافته بر مبنای توزیع ویبول از اطمینان بیشتری نسبت به سایر توزیع ها برخوردار بود (2/322- = AICc برای فالاریس، 8/262- =AICc برای تاج خروس ریشه قرمز و 9/507- =AICc برای تاج خروس خوابیده). مقدار پارامتر شکل نشان داد که توزیع پتانسیل آب پایه هر سه جمعیت بذری چوله به راست است (93/0=λ برای فالاریس، 75/1=λ برای تاج خروس ریشه قرمز و 21/2=λ برای تاج خروس خوابیده). بر مبنای مدل هیدروتایم ویبول، ثابت هیدروتایم و آستانه پتانسیل آب برای شروع جوانه زنی بذر فالاریس به ترتیب 64/106 مگاپاسکال ساعت و 52/1- مگاپاسکال، برای تاج خروس ریشه قرمز به ترتیب 47/20 مگاپاسکال ساعت و 86/0- مگاپاسکال و برای تاج خروس خوابیده به ترتیب 61/76 مگاپاسکال ساعت و 07/1- مگاپاسکال برآورد شد. با توجه به انعطافپذیری توزیع ویبول، این مدل امکان پیش بینی دقیق جوانه زنی و نیز تعیین توزیع پتانسیل آب پایه را فراهم میآورد.
